摘 要:目的探讨miR-124-3p是否通过调节上皮间质转化(EMT)抑制人胃癌细胞SGC-7901的迁移和增殖。方法培养人胃上皮细胞(GES-1)和人胃癌细胞(SGC-7901、MGC-803、BGC-823);使用RT-PCR法检测人胃上皮细胞和人胃癌细胞中miR-124-3p的表达差异;对SGC-7901细胞中miR-124-3p进行上调和下调,上调组分为Control组(Lip2000处理SGC-7901细胞)、mimic-NC组(Lip2000+miR-124-3p拟似物mimic转染SGC-7901细胞)、mimic-miR-124-3p组(Lip2000+mimic-miR-124-3p共转染SGC-7901细胞),下调组分为Control组(Lip2000处理SGC-7901细胞)、inhibitor-NC组(Lip2000+miR-124-3p拟似物inhibitor转染SGC-7901细胞)、si-miR-124-3p组(Lip2000+inhibitor-miR-124-3p共转染SGC-7901细胞)。采用MTT法检测细胞增殖,RT-PCR法检测细胞mRNA表达,Transwell检测细胞迁移,Western blot检测上皮间质转化相关蛋白vimentin表达。结果与GES-1细胞相比,miR-124-3p在SGC-7901中表达显著下降(P<0.05);上调SGC-7901细胞中miR-124-3p后vimentin表达下降,与Control组相比,mimic-miR-124-3p组SGC-7901细胞增殖和迁移能力降低(P<0.05),mimic-NC组无明显变化(P>0.05);下调SGC-7901细胞中miR-124-3p后vimentin表达增加(P<0.05),与Control组相比,inhibitor-miR-124-3p组SGC-7901细胞增殖和迁移能力增强(P<0.05),inhibitor-NC组无明显变化(P>0.05)。结论在胃癌SGC-7901细胞中miR-124-3p低表达可能是激活肿瘤细胞EMT的原因,并促使肿瘤细胞增殖和迁移。Objective To investigate whether miR-124-3 p inhibit the migration and proliferation of human gastric cancer cell SGC-7901 by regulating epithelial-mesenchymal transition(EMT).Methods Cultured human gastric epithelial cells(GES-1)and human gastric cancer cells(SGC-7901,MGC-803,BGC-823)were used as the target cells.RT-PCR was used to detect the expression difference of miR-124-3 p in human gastric epithelial cells and human gastric cancer cells.miR-124-3 p in SGC-7901 cells was up-regulated and down-regulated respectively.Up-regulated group was divided into control group(SGC-7901 cells treated with Lip2000),mimic-NC group(Lip2000+miR-124-3 p mimic mimics transfected SGC-7901 cells),mimic-miR-124-3 p group(Lip2000+mimicmiR-124-3 p co-transfected SGC-7901 cells).Down-regulated group was divided into control group(SGC-7901 cells treated with Lip2000),inhibitor-NC group(Lip2000+miR-124-3 p inhibitor mimics transfected SGC-7901 cells),simiR-124-3 p group(Lip2000+inhibitor-miR-124-3 p co-transfected SGC-7901 cells).MTT method was used to detect cell proliferation.RT-PCR method was used to detect cell mRNA expression.Transwell was used to detect cell migration.Western blot was used to detect the expression of epithelial-mesenchymal transition related protein vimentin.Results Compared with GES-1 cells,the expression of miR-124-3 p in SGC-7901 cells was decreased significantly(P<0.05).The expression of vimentin protein in SGC-7901 cells decreased after up-regulation of miR-124-3 p.Compared with the control group,the migration and proliferation ability of SGC-7901 cells in the mimic-miR-124-3 p group decreased significantly(P<0.05),but there was no significant change in mimic-NC group(P>0.05).The expression of vimentin protein in SGC-7901 cells increased after down-regulation of miR-124-3 p(P<0.05).Compared with the control group,the migration and proliferation ability of SGC-7901 cells in the inhibitor-miR-124-3 p group increased significantly(P<0.05),but there was no significant change in the inhibitor-NC group(P>0.05).Co
