摘　　要：目的探讨尾型同源盒转录因子2(CDX2)作为大肠癌(CRC)潜在生物标志物的价值。方法选择2017年7月~2019年3月在我院就诊的96例CRC患者临床资料和组织样本,采用实时荧光定量PCR(qRT-PCR)检测CRC患者癌组织与正常癌旁组织中CDX2的表达水平;通过受试者工作特征曲线(Receiver operating characteristic curve,ROC曲线)评估CDX2对CRC的诊断价值;对患者进行随访并统计CDX2低表达和高表达CRC患者的生存情况。结果CRC组织和正常癌旁组织中CDX2表达水平分别为0.360±0.108、0.744±0.144,两者比较差异有统计学意义(P<0.0001);ROC曲线结果显示AUC=0.969,敏感度为95.00%,特异度为92.00%;在总生存期(OS)和无进展生存期(PFS)分析中,不同CDX2表达水平的CRC患者生存曲线均存在统计学差异(P=0.043、0.027)。结论CDX2在CRC组织中表达水平明显下降,对CRC具有较高的诊断价值,不同CDX2表达水平的CRC患者生存差异显著,CDX2可能成为CRC预后评估的潜在生物标志物。Objective To explore the value of caudal-type homeobox transcription factor 2(CDX2)as a potential biomarker for colorectal cancer(CRC).Methods The clinical data and tissue samples of 96 CRC patients in our hospital from Jul 2017 to Mar 2019 were enrolled.q RT-PCR was used to detect the expression level of CDX2 in cancer tissues and normal adjacent tissues of CRC patients.The receiver operating characteristic curve(ROC curve)was used to assess the value of CDX2 in the diagnosis of CRC.Patients were followed up,and the survival of patients with low and high expression of CDX2 was analyzed.Results The expression level of CDX2 in CRC tissue and normal adjacent tissue was 0.360±0.108 and 0.744±0.144 respectively,and the difference was statistically significant(P<0.0001).The ROC curve showed that AUC=0.969,with sensitivity of 95.00%and specificity of 92.00%.In the analysis of overall survival and progression-free survival,the survival curves of patients with different expression level of CDX2 were statistically different(P=0.043,0.027,respectively).Conclusion The expression level of CDX2 in CRC tissue decrease significantly,which has high diagnostic value for CRC.The survival of patients with different expression level of CDX2 is significantly different.CDX2 may be used as a potential biomarker for prognosis evaluation of CRC.
