摘  要:目的通过围产期腹腔注射地卓西平马来酸盐(MK-801)建立不同性别的精神分裂症发育小鼠模型,探讨利培酮(RIS)治疗对不同性别精神分裂症发育模型小鼠行为学的影响。方法将60只出生P 7 d(P7 d)的C57BL/6小鼠按性别分为两大组,每大组又随机分为对照组、模型组和RIS治疗组,每组10只。模型组和RIS治疗组于P7 d接受0.6 mg·kg^(-1)的MK-801腹腔注射,每日1次,连续注射5 d,对照组以生理盐水(NS)平行处理,注射体积为10 mL·kg^(-1)。RIS治疗组于P45 d接受0.3 mg·kg^(-1)RIS腹腔注射,对照组和模型组小鼠以NS平行处理,每日1次,连续注射15 d,P60 d开始进行旷场实验和社会交往实验。结果与对照组比较,雄性和雌性模型组小鼠自发活动总路程显著增加(P<0.05),社会交往时间和非攻击性行为百分比均显著减少(P<0.01);雄性和雌性RIS治疗组小鼠自发活动总路程显著增加(P<0.05);与模型组比较,雄性和雌性RIS治疗组小鼠自发活动总路程显著减少(P<0.05),社会交往时间和非攻击性行为百分比均显著增加(P<0.01,P<0.05);各组组内雄性小鼠与雌性比较,自发活动总路程和非攻击性行为百分比均显著增加(P<0.05)。结论围产期MK-801重复处理可引起雌性和雄性小鼠成年后的自发活动增加及社交功能损害,且行为学改变存在性别差异,RIS治疗可不同程度地改善MK-801诱导的雄性和雌性小鼠上述行为学异常。Objective To determine the effect of risperdone(RIS)on the behaviors of schizophrenia developmental model mice,and to establish a mouse model of schizophrenia in different genders established by perinatal intraperitoneal injection of dizocilpine maleate(MK-801).Methods Sixty male and female C57BL/6 mice(7 days after birth,P7 d)were divided into 2 groups according to sex.Each group was randomly divided into a control group,a model group and a RIS treatment group(10 rats in each group).The model group and the RIS group received 0.6 mg·kg^(-1)intraperitoneal injection of MK-801 on day P7,once daily for 5 consecutive days,while the control group was given normal saline in parallel with an injection volume of 10 mL·kg^(-1).The RIS group received intraperitoneal injection of 0.3 mg·kg^(-1)RIS on day P45,while the mice in the control group and the model group were treated with normal saline in parallel,once daily for 15 consecutive days.Open field tests and social interaction tests were conducted on day P60.Results Compared with the control group,the total distance of spontaneous activities of the male or female model group was significantly increased(P<0.05),and the time of social interaction and the percentage of non-aggressive behaviors were significantly reduced(P<0.01).The total distance of spontaneous activities of the mice in the male or female RIS treatment group was significant increased(P<0.05).Compared with the model group,the total distance of spontaneous activities of the male or female RIS treatment group mice was significantly reduced(P<0.05),but the time of social interaction and the percentage of non-aggressive behaviors were significantly increased(P<0.01,P<0.05).When comparing male mice in each group with females,the total distance of spontaneous activities and the percentage of non-aggressive behaviors were increased(P<0.05).Conclusion Repeated treatment with MK-801 during the perinatal period causes increased spontaneous activity and impairment of social function in adult female and male mice,wit
