摘 要:目的研究DRD4/cAMP/PKA途径在酒精依赖大鼠海马区的表达情况。方法雄性SD大鼠随机数字表法分为对照组(n=8)和模型组(n=8),双瓶连续饮用10%酒精制备酒精依赖模型,记录建模期间大鼠的体重变化、酒精摄入量、酒精偏好,通过高架十字迷宫和酒精戒断综合征评分记录大鼠的行为学变化;建模成功后,采用qPCR法检测大鼠海马组织中多巴胺D4受体(DRD4)、腺苷酸环化酶(AC)、cAMP依赖性蛋白激酶A(PKA)、cAMP反应元件结合蛋白(CREB)等指标的mRNA的水平。结果大鼠最后7 d酒精摄入量(7.11±0.36)g/(kg·24 h)、酒精偏好(57.30±1.86)%趋于稳定,即成功制备SD大鼠酒精依赖模型。与对照组比较,模型组DRD4 mRNA相对表达水平(2.75±0.26)显著升高(P<0.001),AC mRNA相对表达水平(0.50±0.02)、PKA mRNA相对表达水平(0.30±0.01)、CREB mRNA相对表达水平(0.47±0.07)显著下降(P<0.01)。结论DRD4可能通过调控cAMP/PKA通路在酒精依赖中发挥重要作用。Objective To explore the expression of DRD4/cAMP/PKA pathway in hippocampus of alcohol-dependent rats.Methods Male SD rats drank two bottles of 10%alcohol continuously to established the model of alcohol dependence,and were randomly divided into a control group(n=8)and a model group(n=8)through Excel software.The change of weight,ethanol intake,and ethanol preference of the rats during the modeling period were recorded.Then,the behavioral changes of the rats through the elevated plus-maze test and evaluation of ethanol withdrawal syndrome were recorded.Intentionally,relative indicators such as DRD4,adenylate cyclase(AC),cAMP-dependent protein kinase A(PKA),and cAMP-response element binding protein(CREB)in the hippocampus were measured using RT-qPCR.Results The alcohol intake(7.11±0.36)g/(kg·24 h)and alcohol preference(57.30±1.86)%of the rats in the last 7 days became stable,which means that the SD rat alcohol dependence model was successfully prepared.Compared with the control group,the expression of DRD4 mRNA(2.75±0.26)in the model group was significantly increased(P<0.001),while the expression levels of AC mRNA(0.50±0.02),PKA mRNA(0.30±0.01)and CREB mRNA(0.47±0.07)were significantly decreased(P<0.01).Conclusion The data summarized here suggests that DRD4 may play an important role in alcohol dependence by regulating the cAMP/PKA pathway.
