摘 要:离子交换层析分离混合氨基酸是应用化学实验课程中一个重要的综合实验。为了让学生更好的掌握离子交换技术的基本理论及实验操作,对阳离子交换树脂的预处理方法进行了改进,学生分组试验对比研究了2种方法下再生的树脂分别重复利用3次的分离效果。试验结果表明采用改进方法预处理和再生重复利用的732型阳离子交换树脂,对比于传统方法,混合氨基酸(Asp、Arg)的分离效果均得以优化,其对2种氨基酸的洗脱都较为集中,拖尾也较短,节省了实验时间和洗脱液用量,实验教学效果显著提高。The separation of mixed amino acids by ion-exchange chromatography is a very important comprehensive experiment for applied chemistry.In order to have a better understanding of the basic theory and experimental operation in ion-exchange technology,an improved method for cation-exchange resin pretreatment has been put forward.The regenerated resin under the improved method and the traditional method were reused three times to compare the amino acid separation differences in groups.Experimental results demonstrated that the 732 cation-exchange resins pretreated by the improved method displayed more optimal effects for the mixed amino acids(Asp & Arg) separation compared with that pretreated by the traditional method.The improved method made the amino acids elution more concentrated and less tailing,reducing the experimental time-consuming and reagent consumption.The experimental teaching effects have been improved obviously.
