宽胸理肺汤合三子养亲汤对COPD大鼠炎症因子、氧化应激及凋亡相关蛋白的影响

摘　　要：目的观察宽胸理肺汤合三子养亲汤对慢性阻塞性肺疾病(COPD)大鼠炎症因子、氧化应激指标、凋亡相关蛋白表达的影响。方法取7只健康雄性Wistar大鼠作为正常组,另取35只雄性Wistar大鼠采用熏香烟加气管注内毒素法建立COPD模型。将建模成功大鼠随机分成COPD组、中药低剂量组、中药中剂量组、中药高剂量组、乙酰半胱氨酸组,每组7只。中药高、中、低剂量组分别按照5,2.5,1.25 g生药/kg的剂量给予宽胸理肺汤合三子养亲汤灌胃,乙酰半胱氨酸组给予0.8 g/kg乙酰半胱氨酸灌胃,正常组和COPD组分别给予等体积0.5%羧甲基纤维素钠灌胃,1次/d,持续28 d。末次灌胃后24 h,心脏取血,采用ELISA法检测血清炎症因子[白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)]及氧化应激指标[丙二醛(MDA)、超氧化物歧化酶(SOD)]水平,HE染色观察大鼠肺组织病理表现,Western blot法检测大鼠肺组织中Bcl-2、Bax、Caspase-3、Caspase-9蛋白表达情况。结果与正常组比较,COPD组大鼠血清IL-6、IL-8、TNF-α、MDA水平及肺组织中Bax、Caspase-3、Caspase-9蛋白表达量均显著升高(P均<0.05),血清SOD水平及肺组织中Bcl-2蛋白表达量均显著降低(P均<0.05);HE染色示大鼠小气道管腔明显狭窄,结缔组织增生,支气管管壁增厚,黏液细胞增多,部分可见黏液堵塞气道,肺泡结构紊乱,上皮细胞不同程度皱缩破坏,管腔及周围血管可见中性粒细胞聚集。与COPD组比较,中药各组及乙酰半胱氨酸组血清IL-6、IL-8、TNF-α、MDA水平及肺组织中Bax、Caspase-3、Caspase-9蛋白表达量均显著降低(P均<0.05),血清SOD水平及肺组织中Bcl-2蛋白表达量均显著升高(P均<0.05);HE染色示气道上皮细胞脱落、炎症细胞浸润、杯状细胞增多以及肺气肿形成等现象均有不同程度减轻。结论宽胸理肺汤合三子养亲汤可以明显改善COPD大鼠的肺组织病理变化,其机制可能与�Objective It is to observe the effects of Kuanxiong Lifei Decoction combined with Sanzi Yangqin Decoction on inflammatory factors,oxidative stress and apoptosis related protein expression in rats with chronic obstructive pulmonary disease(COPD).Methods Seven healthy male Wistar rats were selected as the normal group,and another 35 male Wistar rats were induced by smoking cigarettes and injecting endotoxin into the air tube to establish COPD models.The successfully modeled rats were randomly divided into COPD group,low-dose Chinese medicine group,medium-dose Chinese medicine group,high-dose Chinese medicine group,and acetylcysteine group,with 7 rats in each group.The high,medium and low dose groups of Chinese medicine were given Kuanxiong Lifei Decoction plus Sanzi Yangqin Decoction at the doses of 5,2.5,1.25 g crude drug/kg respectively,and 0.8 g/kg acetylcysteine was given to the acetylcysteine group,the normal group and the COPD group were given an equal volume of 0.5%sodium carboxymethyl cellulose by gavage,all once a day,for 28 days.24 hours after the last gavage,the blood was taken from the heart,and the levels of serum inflammatory factors[interleukin-6(IL-6),interleukin-8(IL-8),tumor necrosis factor-α(TNF-α)]and oxidative stress indexes[malondialdehyde(MDA),superoxide dismutase(SOD)]were detected by ELISA method,the pathological manifestations of rat lung tissue were observed by HE staining,the expression of Bcl-2,Bax,Caspase-3 and Caspase-9 protein were detected by Western blot method.Results Compared with the normal group,the levels of serum IL-6,IL-8,TNF-α,MDA and the protein expression of Bax,Caspase-3 and Caspase-9 in the lung tissue of the COPD group were significantly increased(all P<0.05),the level of serum SOD and Bcl-2 protein expression in lung tissue were significantly reduced(all P<0.05);HE staining showed that the small airway lumen of rats was significantly narrowed,connective tissue was proliferated,bronchial tube wall was thickened,and mucous cells was increased,part of the mucus can bl
