摘 要:目的探究槲皮素通过调控高尔基应激改善脂多糖(LPS)诱导小鼠副鼻窦炎的机制。方法将30只实验小鼠随机分为正常组、模型组和治疗组,使用1 mg/L的LPS诱导小鼠形成副鼻窦炎模型,连续两周通过鼻腔滴入法将LPS滴入小鼠鼻腔内,同时治疗组每天通过灌胃法给予100 mg/kg的槲皮素;通过HE染色观察小鼠鼻黏膜的病理学改变,取小鼠鼻黏膜组织,采用Western blot法检测高尔基体蛋白GM130、高尔基体蛋白GOLPH3和半胱氨酸天冬氨酸蛋白酶3(Caspase3)表达,ELISA法检测炎症因子肿瘤坏死因子-α(TNF-α)和白介素-6(IL-6)含量。结果与模型组比较,治疗组和正常组小鼠的鼻窦黏膜的病理指数评分降低,差异有统计学意义(P<0.05);与正常组比较,模型组小鼠鼻黏膜组织中的GOLPH3和Caspase3表达增加,而GM130蛋白表达减少,差异有统计学意义(P<0.05);与模型组比较,治疗组GOLPH3和Caspase3表达减少,GM130表达增加,差异有统计学意义(P<0.05);与模型组比较,治疗组小鼠鼻黏膜组织的内炎性因子TNF-α和IL-6含量减少,差异有统计学意义(P<0.05)。结论槲皮素可通过调控高尔基应激抑制LPS诱导的小鼠副鼻窦炎应激反应和炎症反应。Objective To explore the mechanism of quercetin to ameliorate LPS-induced paranasal sinusitis in mice through regulating Golgi stress.Methods Thirty experimental mice were randomly divided into 3 groups:normal group,model group and treatment group.The mice were induced to form a paranasal sinusitis model with 1μg/mL LPS.For two consecutive weeks,LPS was instilled into the mice by nasal instillation and the treatment group was given 100 mg/kg of quercetin daily by gavage.HE staining was adopted to observe the pathological changes of mouse nasal mucosa;western blot was adopted to detect the expression of Golgi protein GM130,GOLPH3,and Caspase3;ELISA method was adopted to detect the contents of inflammatory factors TNF-αand IL-6.Results Compared with model group,the pathological index score of the treatment group was higher than that of the normal group and the difference was statistically significant(P<0.05);compared with the normal group,the expression of GOLPH3 and Caspase3 in the nasal mucosa of the model group increased,while the expression of GM130 decreased and the differences were statistically significant(P<0.05);compared with the model group,the expression of GOLPH3 and Caspase3 in the nasal mucosa of the treatment group decreased,while the expression of GM130 increased and the difference was statistically significant(P<0.05);compared with the model group,the contents of TNF-αand IL-6 in the nasal mucosa of the treatment group decreased and the difference was statistically significant(P<0.05).Conclusion Quercetin can inhibit LPS-induced paranasal sinusitis and inflammation in mice by regulating Golgi stress.
