摘 要:目的探讨AGGF1、FOXC2和ATG5与非小细胞肺癌(NSCLC)的相关性,并在其浸润、转移中的可能作用。方法收集2009年2月—2014年5月本院收治的189例NSCLC患者的手术标本,将NSCLC的肿瘤组织作为研究组,并取距肿块>5.0 cm处的正常肺组织作为对照组,采用免疫组织化学染色分别检测AGGF1、FOXC2和ATG5三种蛋白在两组中的表达,并对临床病理数据进行统计学分析和生存分析。结果AGGF1、FOXC2和ATG5蛋白在研究组和对照组中的阳性表达率分别为62.8%(118/188)和30.9%(51/165)、33.0%(62/188)和18.2%(30/165)及52.1%(98/188)和23.6%(39/165),有显著性差异(P<0.05);AGGF1蛋白的表达与NSCLC的TNM分期、分化程度、血管侵犯及淋巴结转移之间差异均有统计学意义(P<0.05);FOXC2蛋白的表达与NSCLC的TNM分期及淋巴结转移之间有显著性差异(P<0.05);ATG5蛋白表达与NSCLC的TNM分期及血管侵犯之间有显著性差异(P<0.05)。Spearman法显示AGGF1、FOXC2和ATG5三种蛋白之间均两两呈正相关关系。Kaplan-Meier法显示上述三种蛋白表达阳性组与阴性组的术后生存率之间,差异均有统计学意义(P<0.05)。Cox多因素分析显示,在NSCLC中AGGF1蛋白的表达、血管侵犯及TNM分期是独立的预后因素。结论AGGF1、FOXC2和ATG5在NSCLC中均呈明显高表达水平,可能三者都参与了NSCLC的侵袭、转移,但是仅AGGF1可能对预测NSCLC进展和预后有重要意义。Objective To investigate the correlation between expression of AGGF1,FOXC2 and ATG5 and non-small cell lung cancer(NSCLC),and their possible roles in invasion and metastasis.Methods Surgical specimen from 189 patients with NSCLC those who admitted to our hospital from February 2009 to May 2014 were collected.The tumor tissues of NSCLC were enrolled as the research group,and normal lung tissues collected at more than 5.0cm from the tumor were taken as the control group.Expression of AGGF1,FOXC2 and ATG5 proteins were detected by immunohistochemistry in the above experimental group and control group.The clinicopathological data were statistically analyzed and survival analysis was performed.Results The positive rate of AGGF1,FOXC2 and ATG5 protein in NSCLC tissues were 62.8%(118/188),30.9%(51/165),and 33.0%(62/188)respectively,and those in the control lung tissues were 18.2%(30/165),52.1%(98/188)and 23.6%(39/165),respectively.There were significant differences between the two groups(P<0.05).There were statistically significant differences between AGGF1 protein expression and TNM stage,degree of differentiation,vascular invasion and lymph node metastasis of NSCLC(P<0.05).FOXC2 protein expression was significantly different in TNM staging and lymph node metastasis in NSCLC(P<0.05).ATG5 protein expression was significantly different in TNM staging and vascular invasion in NSCLC(P<0.05).Spearman analysis showed that AGGF1,FOXC2 and ATG5 were positively correlated with each other(P<0.05).And Kaplan-Meier survival analysis showed that the overall survival rate of NSCLC patients with positive expression of AGGF1,FOXC2 and ATG5 was significantly different from those of NSCLC patients in the negative expression group(P<0.05).COX regression analysis showed that the positive expression of AGGF1 protein,TNM stages and vascular invasion were the independent prognostic factors of NSCLC.Conclusions AGGF1,FOXC2 and ATG5 were highly expressed in NSCLC,and the expression of them might be involved in the invasion and metastasis of NS
