摘 要:目的观察联合应用不同的神经因子对重型脑外伤大鼠神经细胞的影响,探索外源性神经因子治疗重型脑外伤的合适策略及机制。方法 48只SD大鼠随机分为神经生长因子组、碱性成纤维细胞生长因子组、联合组和对照组。脑外伤模型参照改良Feeney法制作,造模1 d后,对照组注射生理盐水,实验各组分别由脑室途径注入神经因子。采用免疫组化法、免疫荧光法比较各组大鼠脑内相应区域阳性细胞的表达。结果在造模3、7、14 d后神经生长因子组、碱性成纤维细胞生长因子组、联合组神经元、神经胶质细胞数量明显多于对照组,差异有统计学意义(P<0.05);同时神经生长因子组、碱性成纤维细胞生长因子组神经元、神经胶质细胞明显少于联合组(P<0.05)。结论联合应用外源性神经生长因子和碱性成纤维细胞生长因子较单用更能促进神经细胞的增殖。Objective To explore the combined use of different nerve factors on severe brain injury after the impact of nerve cells,explore exogenous nerve factors in the treatment of severe brain injury appropriate strategies and mechanisms.Methods Forty-eight SD rats were randomly divided into nerve growth factor group,basic fibroblast growth factor group,combined group and control group.The brain injury model was made according to the modified Feeney method.One day after modeling,normal saline was injected into the control group,and neural factors were injected into each group through the ventricular pathway.The expressions of positive cells in the corresponding regions in the brain of rats in each group were compared by immunohistochemistry and immunofluorescence staining.Results The numbers of neurons and glial cells in nerve growth factor group,basic fibroblast growth factor group and combined group were significantly more than those in the control group 3 d,7 d and 14 d after modeling(P<0.05).At the same time,the numbers of neurons and glial cells in nerve growth factor group,basic fibroblast growth factor group and combined group were significantly less than those in the combined group(P<0.05).Conclusion The combined use of exogenous nerve growth factor and basic fibroblast growth factor can promote the proliferation of nerve cells.
