摘 要:目的观察益气活血方对大鼠心肌梗死后心肌肥大的影响,探究其作用机制。方法采取结扎大鼠冠状动脉的方法建立心肌梗死动物模型,将其随机分为模型组、益气活血药组、酒石酸美托洛尔组,另设假手术组,每组9只,术后第二天开始灌胃,干预治疗7天后,采用小动物超声检测大鼠心脏结构和功能的变化,苏木精—伊红(hematoxylin-eosin,HE)染色法观察梗死边缘区心肌细胞形态学改变,计算心肌细胞横截面积、直径和周长,免疫组织化学法观察梗死边缘区心房钠尿肽(atrial natriuretic peptide,ANP)的表达,蛋白质免疫印迹(western blot,WB)法检测梗死边缘区ANP、β肌球蛋白重链(β-mysion heavy chain,β-MHC)、细胞外信号调节激酶1/2(extracellular signal-regulated kinase1/2,ERK1/2)、磷酸化细胞外信号调节激酶1/2(phosphorylation extracellular signal-regulated kinase1/2,p-ERK1/2)的蛋白表达情况。结果(1)模型组大鼠左心室射血分数(left venteicular ejection fraction,LVEF)、左室短轴缩短率(left ventricular shortening fraction,LVFS)均低于假手术组,左室收缩末期内径(left ventricular internal dimension systole,LVIDs)、左室舒张末期内径(left ventricular end diastolic diameter,LVIDd)均高于假手术组(P<0.05);两用药组LVEF、LVFS均高于模型组,LVIDs、LVIDd均低于模型组(P<0.05)。(2)与假手术组相比,模型组心肌细胞横截面积、直径和周长显著升高;与模型组相比,两用药组三者均明显降低(P<0.05)。(3)Western Blot结果显示,模型组ANP、β-MHC、p-ERK1/2相较假手术组均显著升高;用药组三者相较模型组均显著降低(P<0.05)。ERK1/2蛋白在各组间无统计学差异(P>0.05)。结论益气活血方能够改善大鼠心肌梗死后心肌肥大的程度,其机制可能与抑制ERK1/2信号通路有关。Objective To observe the effects of Yiqi Huoxue formula on myocardial hypertrophy after myocardial infarction in rats,and to explore its mechanism.Methods The animal model of myocardial infarction was established by ligating the coronary artery in rats.They were randomly divided into the model group,the Yiqi Huoxue formula group and the metoprolol tartrate group,meantime the sham operation group was set up,with 9 rats in each group.Gavage was started the next day after surgery.7 days after intervention treatment,the structural and functional changes of rat heart were detected by small animal ultrasound.The hematoxylin-eosin staining method was used to observe the morphological changes of myocardial cells in the edge area of infarction.And the cross-sectional area,diameter and circumference of cardiomyocytes were calculated.The expression of atrial natriuretic peptide(ANP)at the edge of infarction was observed by immunohistochemistry.And western blot method was used to detect the protein expression levels of ANP,β-mysion heavy chain(β-MHC),and extracellular signal-regulated kinase1/2 in the marginal zone of infarction.Results(1)The left venteicular ejection fraction(LVEF)and left ventricular shortening fraction(LVFS)in the model group were lower than those in the sham operation group,and the left ventricular internal dimension systole(LVIDs)and the left ventricular end diastolic diameter(LVIDd)in the model group were higher than those in the sham operation group(P<0.05).LVEF and LVFS in the Yiqi Huoxue formula group and the metoprolol tartrate group were higher than that in the model group,while LVIDs and LVIDd were lower than that in the model group(P<0.05).(2)Compared with the sham operation group,the cross-sectional area,diameter and circumference of myocardial cells in the model group were significantly increased.Compared with the model group,the two medication groups were significantly reduced(P<0.05).(3)Western blot results showed that ANP,β-MHC and p-ERK1/2 in model group were significantly higher than t
