摘 要:阿尔茨海默病(Alzheimer’s disease,AD)与多种癌症的发病存在负相关性,即AD患者合并肿瘤的风险明显降低,反之亦然。虽然二者均为年龄相关性疾病,但其发病呈负相关性的机制却鲜有报道。雌激素相关受体α(estrogen-related receptorα,ERRα)属于核受体NR3B家族,在调节线粒体功能、能量代谢和氧化应激反应等方面发挥着重要的作用。ERRα在AD模型小鼠大脑中表达下调,但在多种类型的肿瘤组织中表达明显上调,提示ERRα在AD与肿瘤的发病过程中发挥着不同的作用。该文综述了近年来研究ERRα在AD与癌症发病中作用的最新进展,为以"整体观念"重新认识AD与癌症发病的相互关系提供理论依据。There is a negative correlation between the incidence of Alzheimer’s disease(AD)and a variety of cancers.That is,the risk of combined tumors in AD patients is significantly reduced,and vice versa.Although AD and tumors are both age-related diseases,their pathogenesis of negative correlation is rarely reported.The estrogen associated receptorα(ERRα)belongs to the nuclear receptor NR3 B family,which is mainly expressed in tissues with high energy demand and plays an important role in mitochondrial function,energy metabolism and oxidative stress.The expression of ERRαwas down-regulated in the brain of AD mouse model,but up-regulated in various types of tumor tissues,suggesting that ERRαplays a different role in AD and the occurrence of tumor.This review summarizes the research progress of ERRαin AD and the incidence of cancer.It is expected to provide a new strategy for the prevention and treatment of AD and cancer while re-recognizing AD and cancer with the"holistic concept".
