摘 要:目的探讨大鼠癫痫持续状态(SE)后海马CA1区大麻素Ⅰ型受体(CB1R)在突触的表达变化。方法取3月龄大鼠分为实验组和对照组,每组6只,建立氯化锂-匹罗卡品大鼠癫痫模型。SE后4周,每组3只大鼠灌注固定,取海马切片,行CB1R免疫组化检测,在显微镜下观察海马CA1区CB1R表达特征;每组剩余3只灌注固定,取海马行CB1R免疫电镜检测。结果CB1R免疫组化DAB显色显示,对照组大鼠海马CA1区CB1R呈点状分布,主要分布在锥体细胞层,SE后4周,实验组大鼠海马CA1区的点、线状分布,较对照明显增加;免疫电镜染色结果显示,对照组大鼠海马CA1区CB1R主要分布在抑制性突触前膜,而在兴奋性突触呈现少量分布。SE后4周,抑制性突触前膜CB1R较对照组明显在增加,而兴奋性突触前膜CB1R表达较对照组无明显变化。结论SE后4周,大鼠海马CA1区CB1R表达增加,并且这种增加主要发生在锥体细胞层抑制性突触前膜。Objective To investigate changes in expression of cannabinoid type I receptor(CB1R)inhippocampal CA1 area of rats with status epilepticus(SE).Methods Three-month-old rats were divided into experimental group and control groupwith 6 rats in each group.Lithium chloride-pilocarpine rat epilepsy model was established.At 4 weeks after SE,3 rats in each group were perfused and fixed.Hippocampal slices were taken for CB1R immunohistochemical detection,and CB1R expression characteristics in hippocampal CA1 area were observed under microscope.The remaining 3 rats in each group were perfused and fixed,and hippocampus were taken for CB1R immunoelectron microscopy.Results CB1R immunohistochemical DAB coloration showed that CB1R in hippocampal CA1 area of control group was distributed in dots,mainly in pyramidal cell layer.At 4 weeks after SE,experimental group was distributed significantly in dots and lines in hippocampal CA1 area than control group.Immunoelectron microscopy showed that CB1R in CA1 area of hippocampus of rats in control group was mainly distributed in inhibitory presynaptic membrane,with a small amount in excitatory synapse.At 4 weeks after SE,compared with control group,inhibitory presynaptic membrane CB1R increased significantly,while excitatory presynaptic membrane CB1R expression did not change significantly.Conclusion 4 weeks after SE,CB1R expression in CA1 area of rat hippocampus increased that it mainly occurred in inhibitory presynaptic membrane of pyramidal cell layer.
