长治地区人群中HLA-B^(*)1502基因频率分析

摘　　要：目的:了解长治地区人群HLA-B*1502的基因频率,为以后研究山西地区该基因频率提供数据,并进一步探讨筛查HLA-B*1502基因的必要性。方法:选取347例需服用卡马西平的患者(糖尿病周围神经痛197例,癫痫78例,三叉神经痛53例,其他神经痛19例)为观察组,100名健康人为对照组。从全血中提取DNA,采用荧光聚合酶链式反应溶解曲线法(以下简称PCR法)和核酸序列测定技术(以下简称测序)对所有样本进行测定。结果:观察组(347个样本)中HLA-B*1502基因的基因频率为0.013,9例(2.59%)为HLA-B*1502基因阳性;对照组(100个样本)中HLA-B*1502基因的基因频率为0.015,3例(3.00%)为HLA-B*1502基因阳性。PCR法与测序法高度一致。结论:长治地区人群中的HLA-B*1502基因频率较中国大部分地区低,但在世界范围内属较高频率人群,对于需服用卡马西平的患者筛查HLA-B*1502基因是有必要的。Objective:To understand the HLA-B*1502 gene frequency of the population in Changzhi area,and to provide data for the future study of the gene frequency in Shanxi Province,and to explore the necessity of screening the HLA-B*1502 gene.Methods:In this study,347 patients(197 patients with diabetic peripheral neuralgia,78 patients with epilepsy,53 patients with trigeminal neuralgia and 19 patients with other neuralgia)who need to take carbamazepine were selected as the experimental group,100 healthy people as the control group.DNA was extracted from the whole blood,and all samples were analyzed by fluorescence polymerase chain reaction dissolution curve method(PCR)and nucleic acid sequencing technology(sequencing).This is to be determined.Results:The gene frequency of HLA-B*1502 gene in the observation group(347 samples)was 0.013,9 cases(2.59%)were positive for HLA-B 1502 gene.The gene frequency of the HLA-B*1502 gene in the control group(100 samples)was 0.015,and 3 cases(3.00%)were positive for the HLA-B 1502 gene.PCR method is highly consistent with sequencing method.Conclusion:The frequency of HLA-B*1502 gene in Changzhi area is lower than that in most parts of China,and it is higher in the world.It is necessary to screen HLA-B*1502 gene for patients who need to take carbamazepine.
