摘  要:目的:观察针刺肺俞、大椎、风门穴对哮喘模型大鼠肺组织炎症区域分子1(FIZZ1)、NOTCH1含量表达的影响,探讨"邵氏五针法"对哮喘气道重塑的作用机制。方法:选取雄性SPF级SD大鼠40只随机分为空白组、模型组、针刺组和艾灸组4组,每组10只。实验第1、8天采用腹腔注射10%复合卵蛋白(OVA)抗原液1mL致敏,从实验的第15天开始,雾化吸入1%OVA溶液激发20min(每日1次,共7次),建立哮喘大鼠模型。针刺组、艾灸组大鼠皆选取肺俞(双)、大椎、风门(双)穴,分别进行针刺和艾灸治疗,治疗后激发。空白组大鼠用0.9%氯化钠溶液代替OVA进行腹腔注射及雾化吸入。末次治疗后摘取包括肺门的右肺中叶肺组织,固定1周后石蜡包埋,制备病理切片,行苏木精-伊红(HE)染色及免疫组织化学染色,观察各组大鼠肺组织形态学改变以及FIZZ1、NOTCH1、α-平滑肌肌动蛋白(α-SMA)在各组大鼠肺组织中的表达,并分析FIZZ1、NOTCH1与α-SMA之间的相关性。结果:与空白组比较,模型组大鼠肺组织中FIZZ1、NOTCH1、α-SMA表达显著增高(P<0.05);与模型组比较,针刺组、艾灸组大鼠肺组织中上述指标表达显著降低(P<0.05),针刺调节炎性蛋白FIZZ1、NOTCH1更为显著(P<0.05)。将研究结果进行相关性分析,FIZZ1、NOTCH1与α-SMA呈正相关性(r=0.899,P<0.01;r=0.889,P<0.01)。结论:"邵氏五针法"可通过下调FIZZ1、NOTCH1与α-SMA的阳性表达,抑制成纤维细胞的分化,减少平滑肌增生,改善哮喘发生发展过程中的气道炎症及气道重塑这一病理环节,这可能是针刺治疗哮喘的作用机制之一。Objective:To observe the effects of acupuncture at Feishu(BL 13),Dazhui(DU 14)and Fengmen(BL12)on the expression of FIZZ1 and NOTCH1 in lung tissue of asthmatic model rats,and to explore the mechanism of‘SHAOs’method of Wuzhen’on airway remodeling of asthma,providing a more scientific theoretical basis for the mechanism study of acupuncture in treating asthma.Methods:Forty male SPF SD rats were randomly divided into four groups in each group:blank group,model Group,acupuncture group and moxibustion group.On the first and eighth days,10%compound OVA antigen solution was injected intraperitoneally for 1 mL sensitization.Starting from the fifteenth day of the experiment,the asthmatic rat model was established by inhaling 1%VA solution for 20 minutes(once a day for 7 times).The treatment group were treated with acupuncture and moxibustion at Feishu(BL 13),Dazhui(DU 14)and Fengmen(BL 12).After treatment,the rats were stimulated.Rats in the blank group were injected intraperitoneally with saline instead of ovalbumin and inhaled by atomization.After the last treatment,the right middle lobe lung tissue including hilum was removed and fixed in 4%paraformaldehyde for one week.Pathological sections were prepared by paraffin embedding,and hematoxylin-eosin(HE)staining and immunohistochemical staining were performed to observe the morphological changes of lung Tissues and the expression of FIZZ1,NOTCH1 andα-SMA in the lung tissues of rats in each group.The correlation between FIZZ1,NOTCH1,α-SMA was analyzed.Results:Compared with the blank group,the expression of FIZZ1,NOTCH1 andα-SMA in the model group was significantly increased(P<0.05).Compared with the model group,the expression of the indexes in the acupuncture and moxibustion groups was significantly decreased(P<0.05),acupuncture regulated inflammatory proteins FIZZ1 and NOTCH1 more significantly(P<0.05).The correlation analysis of the results showed that FIZZ1,NOTCH1 andα-SMA had a strong positive correlation(P<0.01).Conclusion:‘SHAOs’method of Wuzhen’ca
