鸭源性H6N6禽流感病毒对小鼠致病性研究

摘  要:目的:探讨鸭源H6N6亚型禽流感病毒(AIV)是否能跨种间屏障感染哺乳动物小鼠及其基因特点。方法:首先将前期分离的3株鸭源性AIVH6N6,应用基因测序和分析法,比较病毒各基因片段的变化,然后将病毒接种BALB/c小鼠,观察病毒在小鼠复制、感染、病理变化及其组织亲嗜性。结果:基因测序和分析显示,鸭源H6N6亚型AIV的血凝素(HA)裂解位点模式属低致病性病毒,其HA、神经氨酸酶(NA)、PB2等关键基因位点未出现突变,病毒与禽型受体SAα-2,3Gal特异性结合。病毒接种小鼠后,小鼠鼻甲、气管、肺组织出现炎症反应和病毒分离阳性。免疫组化检测病毒NP蛋白显示小鼠鼻甲、气管和支气管黏膜上皮细胞以及肺泡细胞阳性,其中A/DK/KM/1135/2019病毒株复制、感染能力较强。结论:鸭源性AIVH6N6可跨种间屏障传播给哺乳动物,提示有必要持续、密切监测水禽H6N6病毒的进化演变,以防其传播给人。Objective:To investigate whether duck-originated H6 N6 subtype avian influenza virus could cross interspecies barrier from poultry to mammals and its genetic characteristics.Methods:Firstly,three strains of avian influenza virus H6 N6 isolated from ducks were sequenced and the characteristics of gene fragments of H6 N6 strains were analyzed.Secondly,the viruses were inoculated into BALB/c mice to observe the viral replication,infection,pathological changes,and tissue tropism in mice.Results:The HA cleavage site pattern of H6 N6 subtype avian influenza virus from duck belonged to low pathogenicity virus.No mutation occurred in HA,NA,PB2 and other key gene sites,and the viruses specifically bound to avian-like receptor SAα-2,3 Gal.After virus inoculation,inflammation and virus isolation were observed in nasal turbinate,trachea,and lung tissues of mice.The NP of the virus was detected in the epithelial cells of the nasal turbinate,trachea and bronchus and alveolar cells by immunohistochemistry.The A/DK/KM/1135/2019 virus strainhad strong replication and infection ability.Conclusion:Duck-originated H6 N6 subtype avian influenza virus can cross interspecies barrier from poultry to mammals without prior adaption.This study emphasizes the importance of continuous and intense monitoring of the evolution of the H6 N6 virus in aquatic bird.
