摘 要:目的:探讨衰老标记蛋白30(SMP30)在不同月龄大鼠晶状体上皮细胞(LECs)中的表达及其作用。方法:将自然衰老的Wistar大鼠分为1~2月龄组、9~10月龄组及18~19月龄组。采用比色法检测大鼠血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性及丙二醛(MDA)含量,TUNEL法检测大鼠LECs的凋亡情况,间接免疫荧光法检测LECs中SMP30的表达。结果:18~19月龄组血清SOD活性低于1~2月龄组及9~10月龄组,MDA含量和LECs凋亡率高于1~2月龄组及9~10月龄组(均P<0.05);9~10月龄组GSH-PX活性明显低于1~2月龄组(P<0.05),表明大鼠自然衰老模型构建成功。裂隙灯观察3组大鼠晶状体均透明。SMP30在3组大鼠LECs中均呈阳性表达,1~2月龄组、9~10月龄组、18~19月龄组大鼠LECs中SMP30表达的平均荧光强度分别为4.36±1.98、2.73±1.14、0.75±0.39,组间两两比较,差异均有统计学意义(均P<0.05)。结论:随着大鼠月龄增加,LECs凋亡率升高,SMP30在LECs中的表达降低;SMP30可能为LECs的保护因子,在大鼠衰老过程中可能发挥抗LECs凋亡的作用。Objective:To explore the expression and function of senescence marker protein 30(SMP30)in lens epithelial cells(LECs)of different months old rats.Methods:Natural aging Wistar rats were divided into three groups:1-2 months old group,9-10 months old group and 18-19 months old group.The activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)and the content of malondialdehyde(MDA)in serum were determined by colorimetry.Apoptosis of LECs cells was detected by TUNEL assay.The expression of SMP30 in LECs was detected by indirect immunofluorescence assay.Results:Compared with the 1-2 month groupand the 9-10 month group,the serum SOD activity decreased while the MDA content and the apoptotic cells increased in the 18-19 month group(P<0.05);the activity of GSH-PX in the 9-10 month group was significantly lower than that in 1-2 month group(P<0.05),indicating that the natural aging model of rats was successfully constructed.The lens of the three groups of rats were observed to be transparentby slit lamp.SMP30 was positively expressed in LECs in the three groups and the mean fluorescence intensity was 4.36±1.98,2.73±1.14 and 0.75±0.39 in the 1-2 month group,9-10 month group and 18-19 month group,respectively(P<0.05).Conclusion:With the increase of age,the apoptosis rate of LECs increased,and the expression of SMP30 in LECs decreased.SMP30 may be a protective factor of LECs and has an anti-apoptosis effect in the aging process of rats.
