摘  要:目的:通过生物信息技术探讨高迁移率族蛋白B1(HMGB1)促进肺癌进展的潜在关键通路和环节。方法:UALCAN数据库分析HMGB1表达相关的基因和相关基因的生存分析。STRING数据库分析蛋白质-蛋白质相互作用。AnimalTFDB3.0和JASPAR数据库预测转录因子以及结合位点。KEGG数据库分析相关信号通路。结果:生物信息分析结果显示细胞周期蛋白依赖激酶1(CDK1)、H2A组蛋白家族成员Z(H2AFZ)与HMGB1的表达在肺腺癌(LUAD)中呈正相关关系,且CDK1和H2AFZ与LUAD的不良预后有关。预测到SOX2和SP1是CDK1或H2AFZ的转录因子。信号通路分析结果显示Akt和ERK可以调控SOX2的表达。结论:CDK1、H2AFZ可能是HMGB1参与LUAD进展的重要节点,HMGB1-Akt/ERK-SOX2-CDK1/H2AFZ可能是HMGB1促进LUAD进展的关键通路。Objective:To explore the potential signaling pathways and links of high mobility group box 1 protein(HMGB1)promoting the progression of lung cancer by bioinformatics.Methods:The UALCAN database was used to analyze the genes related to HMGB1 expression and the survival analysis of related genes.Analysis of protein-protein interaction was conducted by STRING database.AnimalTFDB3.0 and JASPAR database predicted transcription factors and binding sites.KEGG database analyzed related signal pathways.Results:Bioinformatics analysis showed that cyclin dependent kinase 1(CDK1)or H2 A histone family,member Z(H2 AFZ)were positively correlated with HMGB1 expression in lung adenocarcinoma(LUAD),and CDK1 and H2 AFZ were respectively related to the poor prognosis of LUAD.It was predicted that SOX2 and SP1 were transcription factors of CDK1.The analysis of signal pathways showed that Akt and ERK could regulate the expression of SOX2.Conclusion:CDK1 or H2 AFZ may be an important node for HMGB1 to participate in the progression of LUAD,and HMGB1-Akt/ERK-SOX2-CDK1/H2 AFZ may be a key pathway of HMGB1 promoting the progression of LUAD.
