胃癌组织的LINC01105水平及临床意义

摘　　要：目的探讨胃癌组织的长基因间非蛋白编码RNA 1105(LINC01105)水平及临床意义。方法利用GEPIA在线工具分析LINC01105在癌症基因组图谱(TCGA)中胃腺癌患者癌组织和基因型组织表达数据库(GTEx)正常组织中的水平,采用实时定量PCR(qPCR)检测本院2016年1月至2019年12月手术切除的95例胃癌组织和87例癌旁组织的LINC01105水平,比较胃癌不同临床病理特征的LINC01105水平,Kaplan-Meier Plotter在线数据库分析胃癌组织的LINC01105水平与总生存期(OS)、疾病进展后生存期(PPS)和首次进展后生存期(FP)的关系。结果TCGA数据库中408例胃癌组织的LINC01105中位数水平为0.02,低于211例正常组织的2.45(P<0.05);qPCR结果显示,95例胃癌组织的LINC01105水平为0.636±0.226,低于87例癌旁组织的1.175±0.453(t=10.305,P<0.01)。LINC01105水平与胃癌患者的性别、年龄和分化程度无关(P>0.05),而与肿瘤直径、TNM分期、淋巴结转移和浸润深度有关(P<0.05),其中肿瘤直径较大、TNM分期较晚、淋巴结转移和深浸润组织的LINC01105水平较低。Kaplan-Meier Plotter数据库分析显示LINC01105水平与FP无关(P>0.05),而与OS和PPS有关(P<0.05),其中LINC01105高水平者的中位OS和PPS为93.2和16.7个月,优于低水平者的40.0和10.6个月(P<0.05)。结论胃癌组织中低表达的LINC01105发挥类似抑癌基因的作用,参与了胃癌的恶性进展且与不良预后有关,有作为胃癌诊治靶点的潜能。Objective To investigate the level and clinical significance of long intergenic non-protein coding RNA 1105(LINC01105)in gastric cancer tissues.Methods Levels of LINC01105 in cancer tissues of gastric adenocarcinoma patients in The Cancer Genome Atlas(TCGA)and normal tissues in the Genotype-Tissue Expression(GTEx)project were analyzed with GEPIA online tool.Real-time quantitative PCR(qPCR)was used to detect LINC01105 levels in 95 gastric cancer tissues and 87 adjacent tissues from January 2016 to December 2019.Relationships of LINC01105 level with overall survival(OS),first progression(FP)and post-progression survival(PPS)of gastric cancer were analyzed by Kaplan-Meier Plotter database.Results In TCGA database,the median LINC01105 level in 408 gastric cancer tissues was 0.02,lower than 2.45 in 211 normal tissues(P<0.05).Results of qPCR showed that the LINC01105 level in 95 gastric cancer tissues was 0.636±0.226,lower than 1.175±0.453 in 87 paracancerous tissues(t=10.305,P<0.01).LINC01105 level was not related to gender,age and differentiation degree of gastric cancer patients(P>0.05),but related to tumor diameter,TNM stage,lymph node metastasis and depth of invasion(P<0.05).LINC01105 level was lower in large tumor diameter,advanced TNM stage,lymph node metastasis and deep invasion tissues.Kaplan-Meier Plotter database analysis showed that LINC01105 level was not related to FP(P>0.05),but related to OS and PPS(P<0.05).The median OS and PPS of patients with high LINC01105 level were 93.2 and 16.7 months,better than 40.0 and 10.6 months of patients with low LINC01105 level(P<0.05).Conclusion LINC01105,which is lowly expressed in gastric cancer,plays a role similar to tumor suppressor gene,participates in the malignant progression of gastric cancer and is related to poor prognosis.It has potential as a potential target for the diagnosis and treatment of gastric cancer.
