摘 要:目的:通过检测糖尿病牙周炎大鼠牙周组织中相关炎性因子的表达,探究葡萄籽原花青素提取物(GSPE)对糖尿病牙周炎是否存在影响。方法:选择健康雄性SD大鼠45只随机分为三组:正常对照组(H)、糖尿病牙周炎组(P)和糖尿病牙周炎+GSPE治疗组(G)。采用牙周结扎+高糖高脂饮食+腹腔注射链脲佐菌素建立糖尿病牙周炎大鼠模型,建模后G组给予浓度为100mg/kg GSPE溶液连续灌胃给药40天;检查牙周探诊深度(PD)及附着水平(AL),HE染色观察牙周组织形态,免疫组化染色检测上颌第二磨牙牙龈组织中TNF-α和IL-1β的表达水平。结果:相较H组,P组大鼠牙周组织炎症显著,PD、AL均明显增加,差异具有统计学意义(P<0.05),牙龈组织中TNF-α、IL-1β表达均明显升高,差异具有统计学意义(P<0.05);而G组牙周炎症好转,PD、AL均低于P组,差异具有统计学意义(P<0.05),同时牙龈组织中TNF-α、IL-1β的表达均低于P组,差异具有统计学意义(P<0.05)。结论:GSPE能抑制糖尿病牙周炎大鼠牙龈组织中TNF-α、IL-1β表达,改善其牙周炎症状态及牙槽骨破坏程度,对糖尿病牙周炎具有一定辅助治疗作用。Objective:To investigate the effect of grape seed proanthocyanidin extract(GSPE)on diabetic periodontitis,we detected the expression of related inflammatory factors in the periodontal tissues of diabetic periodontitis rats.Methods:Forty-five male Sprague-Dawley rats were randomly assigned to different groups as follows:control(H),DP(P)and DP+GSPE group(G).The DP model was established by local ligation method+high-fat diet+intraperitoneal injection of STZ.After successful modeling,group G was given GSPE with a concentration of100 mg/kg for continuous gavage administration for 40 days.Hematoxylin-Eosin staining,the depth of periodontal probe,attachment loss,and immunohistochemical staining were performed.Results:Compared with group H,periodontal tissue inflammation was significantly increased in group P,and both PD and AL were significantly increased,the differences were statistically significant(P<0.05),the expression of TNF-αand IL-1βin gingival tissue was significantly increased,and the difference was statistically significant(P<0.05);However,periodontitis improved in group G,PD and AL were lower than those in group P,and the differences were statistically significant(P<0.05),and the expression of TNF-αand IL-1βin gingival tissue was lower than that in P group,and the difference was statistically significant(P<0.05).Conclusion:GSPE can inhibit the expression of TNF-αand IL-1βin gingival tissue of rats with diabetic periodontitis,improve the periodontitis state and the degree of alveolar bone destruction,and have a certain adjuvant effect on diabetic periodontitis.
