摘　　要：目的:通过2-DG作用于人非小细胞肺癌细胞发生的一系列生物学行为改变来探讨线粒体在肿瘤发生发展中的作用。方法:CCK-8检测不同浓度2-DG作用于A549细胞后的抑制率,流式细胞术检测细胞凋亡、细胞周期和线粒体膜电位变化,对2-DG处理后的肿瘤细胞进行线粒体基因组测序。结果:作用48 h后,2-DG的浓度从100μmol/L到10 mmol/L均能明显抑制A549细胞增殖,且随着药物浓度增大,A549细胞抑制率逐渐增大;2-DG作用于A549细胞后可使细胞周期阻滞在S期;2-DG作用于A549细胞后,细胞膜电位从高到低发生改变,说明2-DG可诱导A549细胞的线粒体膜损伤;线粒体基因组测序结果未见明显改变。结论:2-DG可通过诱发线粒体膜损伤进一步诱导细胞周期阻滞以抑制肿瘤细胞增殖。Objective:To investigate the role of mitochondria in tumorigenesis of non-small cell lung cancer through a series of biological behavior changes induced by 2-DG.Methods:CCK-8 was used to detect the inhibition of 2-DG at different concentrations on A549 cells,flow cytometry was used to detect apoptosis,cell cycle and mitochondrial membrane potential,and mitochondrial genome sequencing was performed on tumor cells treated with 2-DG.Results:After 48 h treatment,2-DG concentration from 100μmol/L to 10 mmol/L could significantly inhibit the proliferation of A549 cells,and with the increase of drug concentration,inhibition rate of A549 cells also increased gradually.Different concentrations of 2-DG could significantly inhibit proliferation of tumor cells and arrest the cell cycle in S phase.The cell membrane potential of A549 cells changed from high to low after treatment with different concentrations of 2-DG,indicating that 2-DG can induce mitochondrial membrane damage in A549 cells and the mitochondrial genome sequencing results did not change significantly.Conclusion:2-DG can further induce cell cycle arrest and inhibit tumor growth by inducing mitochondrial membrane damage.
