肝癌组织miR-146b-5p和miR-134的表达下调及临床意义

摘　　要：目的探讨微小RNA(miR)-146b-5p和miR-134在肝癌组织中的表达及其临床病理意义。方法选取2017年6月~2018年12月我院收治的原发性肝癌患者73例,取患者的原发肿瘤组织为观察组,同时选取相对应的癌旁正常组织标本为对照组。采用实时荧光定量PCR法(qRT-PCR)检测癌组织和癌旁正常组织中miR-146b-5p和miR-134的表达水平,并分析两者的表达水平与临床病理特征的相关性。结果miR-146b-5p和miR-134在观察组中的相对表达量均低于对照组(P<0.05);miR-146b-5p的表达下调与肿瘤大小(>5cm)、无静脉浸润和TNM分期(Ⅲ/Ⅳ期)相关(均P<0.05);miR-134的表达下调与肿瘤大小(>5cm)、有肝硬化、TNM分期(Ⅲ/Ⅳ期)和无肿瘤包膜相关(均P<0.05)。结论肝癌组织中,miR-146b-5p和miR-134表达水平下调,可能参与肝癌的发生和发展,与肝癌临床恶性表型密切相关。Objective To explore the expression of microRNA(miR)-146b-5p and miR-134 in liver cancer tissues and their clinical pathological significance.Methods The cancer tissues specimens from the 73 patients with primary liver cancer were enrolled.The corresponding para-cancerous normal tissue specimens were enrolled as controls.The expression levels of miR-146b-5p and miR-134 in cancer tissues and para-cancerous normal tissues were detected by real-time fluorescence quantitative PCR(qRT-PCR).The correlation between their expression levels and clinicopathological features was analyzed.Results The relative expression levels of miR-146b-5p and miR-134 in liver cancer tissues were lower than those in para-cancerous tissues(P<0.05).The down-regulated expression of miR-146b-5p was related to tumor size(longer than 5cm),non-venous infiltration and TNM staging(stageⅢ/Ⅳ)(P<0.05).The down-regulated expression of miR-134 was related to tumor size(longer than 5cm),cirrhosis,TNM staging(stageⅢ/Ⅳ)and non-tumor capsule(P<0.05).Conclusion In liver cancer tissues,the expression levels of miR-146b-5p and miR-134 are down-regulated,which may be involved in the occurrence and development of liver cancer,and are closely related to clinical malignant phenotypes of liver cancer.
