摘  要:目的探讨虾青素(Astaxanthin,AST)调控氧自由基/JNK信号通路对脊髓损伤处神经细胞的保护作用。方法采用高空坠物法的方法制备脊髓损伤小鼠模型。将造模成功的小鼠随机分为SCI组与虾青素组(灌胃给予25 mg/kg的AST),另取正常C57BL/6小鼠作为假手术组。评价各组小鼠后肢功能;检测各组小鼠脊髓含水量;HE染色观测各组小鼠脊髓损伤情况;TUNEL检测各组小鼠脊髓损伤处神经细胞的凋亡情况;免疫荧光检测脊髓组织ROS;Western blot法检测JNK、p-JNK蛋白表达。结果虾青素可以改善SCI小鼠后肢功能,降低脊髓含水量,凋亡细胞数显著降低,抑制线粒体中ROS产生,减轻线粒体肿胀,降低p-JNK蛋白水平。结论虾青素可抑制ROS/JNK通路,改善脊髓损伤中神经细胞的凋亡。Objective To investigate the protective effect of astaxanthin(AST) on neurons after spinal cord injury by regulating oxygen free radical/JNK signaling pathway. Methods The mouse model of spinal cord injury was established by falling objects. The mice were randomly divided into SCI group and astaxanthin group(25 mg/kg AST by gavage), and normal C57 BL/6 mice were selected as sham operation group. The function of hind limbs was evaluated;the water content of spinal cord was detected;the spinal cord injury was observed by HE staining;the apoptosis of nerve cells was detected by TUNEL;the ROS of spinal cord was detected by immunofluorescence;the expression of JNK and p-JNK was detected by Western blot. Results Astaxanthin can improve hindlimb function of SCI mice, reduce spinal cord water content, significantly reduce the number of apoptotic cells, inhibit the production of ROS in mitochondria, reduce mitochondrial swelling, and reduce the level of p-JNK protein. Conclusion Astaxanthin can inhibit ROS/JNK pathway and improve neuronal apoptosis in spinal cord injury.
