摘 要:目的研究FOXN2通过PI3K/AKT信号通路对肾癌增殖、凋亡的影响。方法实验分FOXN2过表达组、空载体对照组、对照组三组;检测FOXN2在人正常肾上皮细胞HK-2和肾癌细胞786-O中的表达;检测FOXN2过表达组、空载体对照组、对照组细胞的增殖情况和凋亡情况;检测FOXN2过表达组、空载体对照组、对照组细胞PI3K、AKT、p-AKT、Bcl-2、Bax、CDK1、CyclinD1蛋白表达水平。结果(1)与人正常肾上皮细胞HK-2相比,人肾癌细胞786-O中的FOXN2在mRNA水平上的表达极显著降低(P<0.01);(2)转染了含有FOXN2的实验组细胞的FOXN2在mRNA水平上的表达量和蛋白水平上的表达量比空载体对照组、对照组细胞的FOXN2表达量极显著增高(P均<0.01)。786-O细胞转染了FOXN2后,增殖能力显著降低(P<0.05);(3)FOXN2过表达组的细胞凋亡率与对照组相比明显增加;(4)FOXN2过表达组p-AKT、凋亡抑制因子Bcl-2、CDK1、CyclinD1蛋白表达降低(P<0.05),Bax的表达明显增高。结论FOXN2可影响肾癌细胞的增殖及凋亡,机制可能通过PI3K/AKT信号通路发挥作用。Objective To study the effect of FOXN2 on proliferation and apoptosis of renal cell carcinoma through PI3 K/AKT signaling pathway.Methods The experiment was divided into three groups:FOXN2 overexpression group,empty vector control group and control group;the expression of FOXN2 in human normal renal epithelial cells HK-2 and renal cancer cells 786-O was detected;the proliferation and apoptosis of FOXN2 overexpression group,empty vector control group and control group were detected;the expression of PI3 K,AKT,p-AKT,Bcl-2,Bax,CDK1 and cyclin in FOXN2 overexpression group,empty vector control group and control group were detected ND1 protein expression level.Results(1)Compared with HK-2 cells,the expression of FOXN2 in 786-O cells was significantly decreased(P<0.01);(2)The expression of FOXN2 in HK-2 cells transfected with FOXN2 was significantly increased(P<0.01).786-O cells transfected with FOXN2 significantly decreased the proliferation ability(P<0.05);(3)The apoptosis rate of FOXN2 overexpression group was significantly increased compared with the control group;(4)The protein expression of p-AKT,Bcl-2,CDK1,CyclinD1 in FOXN2 overexpression group was significantly decreased(P<0.05),and the expression of Bax was significantly increased.Conclusion FOXN2 can affect the proliferation and apoptosis of RCC cells,and the mechanism may play a role through PI3 K/AKT signaling pathway.
