摘 要:目的探讨白细胞相关免疫球蛋白样受体1(LAIR-1)在宫颈癌患者淋巴细胞中的表达及其意义。方法选取2019年1月~2020年11月九江市第三人民医院妇科收治的30例宫颈癌患者纳入宫颈癌组,另选取同期健康体检的30例志愿者纳入健康人组。宫颈癌组患者血液于治疗及术前采集。将宫颈癌组按照随机数字表法分为两个亚组:刺激组15例和非刺激组15例,每组3个复孔,分别加入如下试剂:转化生长因子-β^(+)聚酯-聚羟基脂肪酸(PHA);无PHA、转化生长因子-β。采用Ficoll密度梯度离心法分离外周血单个核细胞(PBMC),在FSC-SSC散点图中圈出淋巴细胞,分析LAIR-1在T细胞亚群CD4^(+)、CD8^(+)、自然杀伤(NK)细胞中的表达水平。结果宫颈癌组CD4^(+)、CD8^(+)T细胞和NK细胞LAIR-1分子表达水平高于健康人组,差异有统计学意义(P<0.05)。刺激组患者CD4^(+)、CD8^(+)、NK细胞中LAIR-1表达水平低于未刺激组,差异有统计学意义(P<0.05)。结论LAIR-1可能参与抑制NK细胞和CTL细胞的杀伤活性等机制,抑制机体的抗肿瘤免疫应答。Objective To investigate the expression and significance of leukocyte-associated immunoglobulin-like receptor 1(LAIR-1)in lymphocytes of patients with cervical cancer.Methods From January 2019 to November 2020,30 patients with cervical cancer in the Department of Gynecology,the Third People′s Hospital of Jiujiang City were included in the cervical cancer group,and 30 volunteers who received physical examinations during the same period were included in the healthy group.The blood of patients in the cervical cancer group was collected before treatment and surgery.The cervical cancer group was divided into two subgroups according to the random number table method:15 cases in the stimulation group and 15 cases in the non-stimulation group.Each group had 3 multiple holes,and the following reagents were added respectively:transforming growth factor-β^(+)polyhydroxyalkanoates(PHA);no PHA,transforming growth factor-β.The Ficoll density gradient centrifugation method was used to separate peripheral blood mononuclear cells(PBMC),the lymphocytes were circled in the FSC-SSC scatter diagram,and the expression levels of LAIR-1 were analyzed in the T cell subsets of CD4^(+),CD8^(+)and natural killer(NK)cells.Results The expression levels of LAIR-1 molecules of CD4^(+),CD8^(+)T cells and NK cells in the cervical cancer group were higher than those in the healthy group,and the differences were statistically significant(P<0.05).The expression levels of LAIR-1 in CD4^(+),CD8^(+)and NK cells in the stimulation group were lower than those in the non-stimulation group,and the differences were statistically significant(P<0.05).Conclusion LAIR-1 may participate in the mechanism of inhibiting the killing activity of NK cells and CTL cells,and inhibit the body′s anti-tumor immune response.
