新疆地区酒精依赖患者HTR3A和HTR3B基因多态性与酒依赖的相关性分析

摘　　要：目的:探讨新疆地区酒精依赖患者5-羟色胺受体3A(HTR3A)和3B(HTR3B)基因与酒精依赖的关系。方法:本研究纳入了符合研究标准的199名酒精依赖男性患者和197名健康男性作为对照,根据国家生物技术中心信息库先前研究确认的HTR3A和HTR3B基因序列,筛选出20个单核苷酸多态性(SNPs),采用SPSS 19.0统计软件包,χ^(2)检验,遗传模型分析和PLINK软件对筛选出的SNP进行关联分析。结果:我们发现在基因型模型中,rs10160548与酒精依赖风险增加有关。对于A等位基因,rs1176722增加了等位基因模型(OR(95%CI)=1.576(1.038-2.394),P=0.033)和相加模型(OR(95%CI)=1.63(1.055-2.517),P=0.028)的风险。结论:新疆地区酒精依赖患者rs1176722、rs10160548和HTR3B基因上的rs2276305单核苷酸多态性与酒精依赖阳性性状有关。Objective:Therefore,the treatment and prevention of alcohol dependence is an urgent public health problem.In this study,the relationship between serotonin receptor 3 A(HTR3 A)and 3 B(HTR3 B)genes and alcohol dependence in a Chinese population was explored.Methods:In this study,199 male patients with alcohol dependence and 197 healthy men who met the research criteria were included as controls.According to the gene sequences of HTR3 A and HTR3 B confirmed in previous studies in the Information Database of National Biotechnology Center,20 single nucleotide polymorphisms(SNPs)were screened out.SPSS 19.0 statistical software,χ^(2)test,genetic model analysis and PLINK software were used to analyze the association of the screened SNPs.Results:We found that rs10160548 was associated with increased alcohol dependence risk in the genotype model.With For the A allele,rs1176722 increased the risk in allele model(OR(95%CI)=1.576(1.038-2.394),P=0.033)and in the additive model(OR(95%CI)=1.63(1.055-2.517),P=0.028).Rs2276305 increased the risk in genotype and dominant model.Conclusions:The rs1176722,rs10160548 and rs2276305 single nucleotide polymorphisms in the HTR3 B gene of alcohol dependent patients in Xinjiang are related to the positive traits of alcohol dependence.
