长链非编码RNA CTB-147C22.8对复发性呼吸道乳头状瘤上皮细胞内蛋白表达的影响

摘　　要：目的探讨长链非编码RNA(long non-coding RNA,lncRNA)CTB-147C22.8 调控下游蛋白对复发性呼吸道乳头状瘤细胞侵袭及细胞周期的影响。方法在前期高通量基因组表达谱芯片分析下游mRNA KLK6基因的基础上,复苏NP69-空白组、NP69-H145组、NP69-H6373组三组细胞系,采用Western blot技术检测转染CTB-147C22.8前、后人上皮细胞中细胞胞内及胞外E-钙黏素(E-cadherin)、β链蛋白(β-catenin)表达量,cyclin E、cyclin D1、cyclin A、CDK2表达量。结果 NP69-H6373组E-cadherin在细胞浆蛋白表达量较NP69-空白组和NP69-H145组增高且均有统计学意义(F=11.192,P=0.09);实验组β-catenin在细胞浆和细胞核表达量与其他两组比表达增高,且有统计学意义(F=2.33,P=0.037;F=2.16,P=0.048);NP69-H6373组cyclin E、CDK2表达量较NP69-空白组和NP69-H145组显著增加,且差异有统计学意义(F=4.132,P=0.043;F=3.981,P=0.037)。结论 lncRNA CTB-147C22.8-KLK6可调控其下游蛋白,E-cadherin、β-catenin及cyclin E、CDK2影响呼吸道乳头状瘤细胞的增殖、侵袭能力。OBJECTIVE To investigate the effect of lncRNA CTB-147C22.8 on the invasion and cell cycle of recurrent respiratory papilloma cells.METHODS Based on the analysis of downstream mRNA KLK6 gene by high-throughput genome expression profiling chip in the early stage,three cell lines of NP69-blank group,NP69-H145 group,and NP69-H6373 group were revived,and the transfection CTB-147C22.8 was detected by Western Blot technology.Before and after the expression of E-cadherin,β-catenin,cyclin E,cyclin D1,cyclin A and CDK2 in human epithelial cells.RESULTS The expression of E-cadherin in the cytoplasmic protein of the NP69-H6373 group was higher than that of the NP69-blank group and the NP69-H145 group and was statistically significant(F=11.192,P=0.09);the expression of β-catenin in the plasma and nucleus was higher in the experimental group than the other two groups,and it was statistically significant(F=2.33,P=0.037;F=2.16,P=0.048).The expression of cyclin E and CDK2 in the NP69-H6373 group was higher than that in the NP69 group.Compared with the blank group and the NP69-H145 group,the expression increased significantly,and the difference was statistically significant(F=4.132,P=0.043;F=3.981,P=0.037).CONCLUSION LncRNA CTB-147C22.8-KLK6 can regulate its downstream proteins:E-cadherin,β-catenin,cyclin E and CDK2,which affect the proliferation and invasion of respiratory papilloma cells.
