摘 要:[目的]探讨槲皮素对软骨肉瘤细胞线粒体功能的影响及作用机制。[方法]软骨肉瘤细胞培养传代后随机分为加药组、激动剂组和空白对照组,加药组加入槲皮素,激动剂组加入槲皮素及740Y-P,空白对照组仅保留肿瘤细胞。干预48 h后检测细胞活性,线粒体膜电位(ΔΨ_(m)),细胞内活性氧(ROS)水平,Akt、p-Akt、Bax、Bcl-2蛋白的表达水平。[结果]加药组细胞活力低于激动剂组(P<0.05)。加药组线粒体ΔΨ_(m)低于激动剂组及空白对照组(P<0.05);加药组ROS含量高于激动剂组及空白对照组(P<0.05)。加药组p-Akt表达水平低于激动剂组及空白对照组(P<0.05)。Bax表达水平加药组高于激动剂组及空白对照组(P<0.05);Bcl-2表达水平加药组低于激动剂组及空白对照组(P<0.05)。[结论]槲皮素能够通过调节PI3K/Akt通路,降低软骨肉瘤细胞线粒体的生理功能,启动线粒体诱导的细胞凋亡程序。[Objective] To explore the effect of quercetin on the mitochondrial function of chondrosarcoma cells and its mechanism.[Method] After culture and passage of chondrosarcoma cells, they were randomly divided into drug group, agonist group and blank control group. The drug group was added with quercetin, the agonist group was added with quercetin and 740 Y-P, while the blank control group had only tumors cell. The cell viability, mitochondrial membrane potential(ΔΨ_(m)), intracellular reactive oxygen species(ROS) and Akt, p-Akt, Bax, and Bcl-2 protein expression levels were detected after 48 hours of intervention. [Results] The drug group had significantly lower cell viability than that of the agonist group(P<0.05). The drug group had significantly lower mitochondrial ΔΨm than the agonist group and the blank control group(P<0.05), whereas the drug group had significantly higher ROS content than the agonist group and the blank control group(P<0.05). In addition, the drug group had significantly lower expression level of p-Akt and Bcl-2, whereas higher Bax expression than the agonist group and the blank control group(P<0.05). [Conclusion] Quercetin does reduce the physiological function of mitochondria of chondrosarcoma cells, and initiate the apoptosis program induced by mitochondria through the PI3 K/Akt pathway.
