摘 要:目的探讨驱动蛋白样DNA结合蛋白(KNSL4)、p21ras在乳腺癌中的表达及与患者淋巴结转移的关系。方法收集82例乳腺癌患者的临床资料和随访资料,依据随访结果将患者分为淋巴结转移组(n=48)、淋巴结未转移组(n=34)。收集乳腺癌患者石蜡包埋的乳腺癌组织及其癌旁组织标本,采用免疫组织化学染色法检测乳腺癌组织及其癌旁组织中KNSL4、p21ras的表达情况,分别采用逆转录聚合酶链反应(RT-PCR)、蛋白质印迹法(Western blot)检测乳腺癌组织中KNSL4、p21ras mRNA和蛋白相对表达量。分析不同临床特征乳腺癌患者乳腺癌组织中KNSL4、p21ras的表达情况,比较淋巴结转移组、淋巴结未转移组乳腺癌患者乳腺癌组织中KNSL4、p21ras mRNA和蛋白相对表达量,采用Spearman相关分析法分析乳腺癌淋巴结转移与KNSL4、p21ras基因表达的相关性。结果乳腺癌组织中KNSL4阳性表达率明显低于癌旁组织,p21ras阳性表达率明显高于癌旁组织,差异均有统计学意义(P﹤0.01)。有淋巴结转移的乳腺癌患者乳腺癌组织中KNSL4阳性表达率明显低于无淋巴结转移的患者,而p21ras阳性表达率明显高于无淋巴结转移的患者,差异均有统计学意义(P﹤0.01)。淋巴结转移组患者乳腺癌组织中KNSL4 mRNA和蛋白相对表达量均明显低于淋巴结未转移组,而p21ras mRNA和蛋白相对表达量均明显高于淋巴结未转移组,差异均有统计学意义(P﹤0.01)。Spearman相关性分析结果显示,乳腺癌淋巴结转移与KNSL4表达量呈负相关,但与p21ras表达量呈正相关(P﹤0.05)。结论乳腺癌淋巴结转移与KNSL4、p21ras基因表达密切相关,KNSL4、p21ras可为乳腺癌的早期诊治提供参考。Objective To investigate the expression of kinesin-like DNA binding protein(KNSL4)and p21ras in breast cancer tissue and its relationship with lymph node metastasis.Method A total of 82 patients with breast cancer were enrolled,and the clinical data and follow-up data were recorded and collected.According to the follow-up outcome,the patients were divided into the lymph node metastasis group(n=48)and the non-lymph node metastasis group(n=34).Paraffin-embedded breast cancer tissues and adjacent tissues were collected,the expression of KNSL4 and p21ras were detected by the immunohistochemical assay based on the tissues.The relative mRNA and protein expression levels of KNSL4 and p21ras were detected by reverse transcription-polymerase chain reaction(RT-PCR)and Western blot,respectively.The expression of KNSL4 and p21ras in breast cancer patients with different clinical features were also analyzed.The relative mRNA and protein expression levels of KNSL4 and p21ras in the lymph node metastasis group and the nonlymph node metastasis group were compared.The correlation between lymph node metastasis of breast cancer and expression of KNSL4 and p21ras genes were analyzed by Spearman correlation analysis.Result The positive rate of KNSL4 in breast cancer tissue was significantly lower than that in adjacent tissue,while the positive rate of p21ras was significantly higher than that in adjacent tissues,the differences were statistically significant(P<0.01).The positive rate of KNSL4 in patients with lymph node metastasis was significantly lower than those without lymph node metastasis,the positive rate of p21ras was significantly higher than patients without lymph node metastasis,the differences were statistically significant(P<0.01).The relative mRNA and protein expression levels of KNSL4 in the lymph node metastasis group were significantly lower than those in the non-lymph node metastasis group,while the relative mRNA and protein expression levels of p21ras were significantly higher than those in the non-lymph node metas
