摘 要:以河北省平泉七沟油松(Pinus tabulaeformis)良种基地育种群体为研究对象,利用EST-SSR引物对来自4处种源的153个油松无性系进行了遗传多样性分析。在12个位点上共检查到62对等位基因,平均等位基因数为5.17;各位点的平均期望和观测杂合度(He和Ho)分别为0.414和0.403,近交系数(Fis)为0.018 6;4个种源群中大窝铺、东陵和七沟3个群体的平均有效等位基因数(Nea)、等位基因丰富度(AR)、观测杂合度(Ho)和期望杂合度(He)基本一致,都略高于宽城群体;平均群体差异系数(Fst)偏低,仅为0.015 6;种源群体间的差异系数(Pairwise Fst)极低,仅在0.009 9~0.071 7之间;NJ树图和主成分分析(PCA)的结果,显示各无性系的亲缘关系较为接近。综上,平泉油松种子园育种群体的遗传多样性水平较丰富,但是无性系之间的亲缘关系较近,群体结构不清晰。In this study the breeding population of Pinus tabulaeformis in Pingquan, Hebei Province was taken as the research object, the genetic diversity of 153 clones from 4 provenances was analyzed using EST-SSR primers. A total of 62 pairs of alleles were detected at 12 loci, and the mean allele number was 5.17. The mean expected and observed heterozygosity(He and Ho) of each point was 0.414 and 0.403 respectively, and the inbreeding coefficient(Fis) was 0.0186. The average number of effective alleles(Nea), allele abundance(AR), observed heterozygosity(Ho) and expected heterozygosity(He) of Dawopu, Dongling and Qigou populations in the4 provenance groups were basically the same, which were slightly higher than that of the Kuancheng population. The mean population difference coefficient(Fst) was low, only 0.0156.The coefficient of difference between provenance populations(Pairwise Fst) was also extremely low, ranging from 0.0099 to 0.0717. The results of NJ tree and principal component analysis(PCA) showed that all clones had close genetic relationships. In conclusion, the genetic diversity of the population in Pinus tabulaeformis variety orchard was abundant, but the clones were closely related and the population structure was unclear.
