摘 要:目的探讨长链非编码RNA前列腺雄激素调节转录因子1(prostate androgen-regulated transcript 1,PART1)对口腔鳞状细胞癌增殖、侵袭和凋亡的影响及其机制。方法采用qRT-PCR方法检测PART1在口腔鳞状细胞癌组织、癌旁正常组织及口腔鳞状细胞癌细胞系中的表达。构建PART1过表达载体,通过转染在口腔鳞状癌细胞系SCC-9和SCC-15细胞中过表达PART1。CCK-8法检测细胞增殖;Transwell实验检测细胞迁移和侵袭能力;Western印迹检测上皮-间充质转化(epithelial-mesenchymal transition,EMT)相关蛋白及PI3K/AKT信号通路蛋白表达变化;流式细胞术检测细胞凋亡率。结果PART1在口腔鳞状细胞癌组织和细胞中呈低表达;过表达PART1显著抑制口腔鳞状细胞癌细胞的增殖、迁移和侵袭;过表达PART1显著上调EMT相关钙黏着蛋白E-cadherin表达,并下调波形蛋白vimentin表达;流式细胞术显示过表达PART1诱导口腔鳞状癌细胞凋亡;过表达PART1显著抑制PI3K和AKT磷酸化水平。结论长链非编码RNA PART1抑制口腔鳞状癌细胞增殖、迁移和侵袭,并促进其凋亡,PI3K/AKT信号通路可能参与调控。Objective To unravel the role and underlying regulatory mechanism of long non-coding RNA prostate androgen-regulated transcript 1(PART1)in oral squamous cell carcinoma(OSCC).Methods The qRT-PCR was used to determine the mRNA level of PART1 in OSCC tissues and cells.PART1 overexpression vector was constructed and transfected into OSCC cell line SCC-9 and SCC-15 cells.Cell proliferation was monitored by CCK-8 assay.Cell migratory and invasive capabilities were assessed by Transwell migration and invasion assays.Western blot was performed to detect the protein levels of epithelial-mesenchymal transition(EMT)markers E-cadherin and vimentin,as well as PI3 K/AKT signaling molecules.Annexin V-FITC/PI staining and flow cytometry were used to assess cell apoptosis.Results PART1 was significantly down-regulated in OSCC tissues and cells.Over-expression of PART1 remarkably suppressed cell proliferation,migration and invasion.PART1 over-expression increased E-cadherin,but decreased vimentin in OSCC cells.Cell apoptotic rate was significantly increased in PART1-over-expresssing cells.These changes were accompanied by the down-regulation of p-PI3 K and p-AKT.Conclusion Long non-coding RNA PART1 suppresses cell proliferation,migration and invasion,and triggers apoptosis in OSCC cells possibly via PI3 K/AKT signaling pathway.
