摘 要:目的采用β-葡萄糖苷酶水解射干苷,并对其酶水解动力学进行初步研究。方法以射干苷的水解转化率为指标,考察各反应因素对射干苷转化率的影响,并计算酶解反应动力学参数。结果射干苷的最佳水解条件为反应时间20 min;溶液pH值为5.0,反应温度65℃,酶浓度100 U·mL^(-1),底物初始浓度0.118 mmol·L^(-1),在此条件下,射干苷水解转化率达到80%以上。该水解反应过程符合单底物Michaelis Menten方程,其中Vmax(最大反应速率)值为20.83 mmol·(L^(-1)·min-1),Km(米氏常数)值为0.21 mmol·L^(-1)。结论采用β-葡萄糖苷酶水解射干苷制备鸢尾黄素可行。Objective Theβ-glucosidase was used to hydrolyze tectoridin and its enzymatic hydrolysis kinetics was studied.Method The effect of various reaction factors on the hydrolysis conversion rate of tectoridin was investigated and the kinetic parameters were calculated.Result The optimal technological parameters of hydrolysis were as the following.The reaction time was 20 min.The pH value was 5.0 and the hydrolysis temperature was 65℃.The enzyme concentration was 100 U·mL^(-1) and the initial substrate was 0.118 mmol·L^(-1).Under these conditions,the hydrolysis conversion rate of ectoridin reached more than 80%.The hydrolysis process conformed to the single substrate Michaelis Menten equation,where the maximum reaction rate(Vm)was 20.83 mmol·(L^(-1)·min-1),and the Km value was 0.21 mmol·L^(-1).Conclusion It is feasible to prepare tectorigenin by hydrolyzing tectoridin withβ-glucosidase.Commercialβ-glucosidase may be a specific hydrolytic enzyme of tectoridin.
