摘 要:目的探究乳铁蛋白(LF)对去卵巢大鼠骨组织中胰岛素样生长因子Ⅰ(IGF-Ⅰ)、胰岛素样生长因子结合蛋白3(IGFBP-3)、胰岛素样生长因子结合蛋白5(IGFBP-5)mRNA的表达及其对该模型骨量的影响,探讨LF对绝经后骨质疏松的作用及可能的分子学机制。方法将70只6月龄雌性SD(Sprague-Dawley)大鼠平均分为7组:假手术组、绝经后骨质疏松模型组、骨质疏松模型雌激素干预组[每周肌内注射0.1 mg/kg]、骨质疏松模型牛血清白蛋白组[口服100 mg/(kg·d)]和不同剂量的骨质疏松模型LF干预组[口服100,1000,2000 mg/(kg·d)]。连续干预6个月后,采用生物光学显微镜拍摄胫骨干骺端的骨组织病理切片,图像分析软件计算各组中骨小梁平均面积所占骨髓腔的比例。结合实时荧光定量聚合酶链式反应(RT-PCR)分析股骨近端骨组织IGF-Ⅰ、IGFBP-3及IGFBP-5 mRNA的表达水平。结果与骨质疏松模型组比较,LF对体质量的影响较小,但在其干预下骨小梁平均面积所占骨髓腔比例增加,差别有统计学意义(P<0.05)。骨组织IGF-Ⅰ及IGFBP-5 mRNA的表达量增加,而IGFBP-3 mRNA的表达量被抑制,差别均有统计学意义(P<0.05)。结论LF可能通过增加骨组织中IGF-Ⅰ及IGFBP-5 mRNA的表达、抑制IGFBP-3 mRNA的表达而减少骨丢失。Objective To investigate the effects of lactoferrin(LF)on the bones in ovariectomized(Ovx)rats and to evaluate the expression of insulin-like growth factor-Ⅰ(IGF-Ⅰ)/insulin-like growth factor binding protein-3(IGFBP-3)/insulin-like growth factor binding protein-5(IGFBP-5)mRNA of the bone tissue.Methods Randomly divided 706-month-old cleaning female Sprague-Dawley rats into 7 groups:the Sham group,the Ovx(Con)group,the Ovx+7β-estradiol[intramuscular injection,0.1 mg/kg,each week]group(E2),the Ovx+bovine serum albumin[peros,100 mg/(kg·d)]group(BSA),the Ovx+Lactoferrin[peros,100 mg/(kg·d)]group(LF1),the Ovx+Lactoferrin[peros,1000 mg/(kg·d)]group(LF2)and the Ovx+Lactoferrin[peros,2000 mg/(kg·d)]group(LF3).By the end of 6 month-treatments,the tibia was obtained and then stained with hematoxylin and eosin(H-E)for the analyses of the mean Trabecular area/Marrow cavity ratio by Image-Pro Plus 6.0.The mRNA levels of IGF-Ⅰand IGFBP-3 and-5 in femur were measured using Real Time PCR.Results LF treatments scarcely influenced the changes in weight,but increased the Trabecular area/Marrow cavity ratio in Ovx rats(P<0.05).Moreover,LF treatments significantly increased the IGF-I/IGFBP-5 mRNA level,but suppressed the level of IGFBP-3 mRNA compared with the untreated Ovx rats(P<0.05).Conclusion Oral administration of LF decreased the bone loss perhaps via increasing the expression of IGF-Ⅰand IGFBP-5 mRNA,while suppressing IGFBP-3 mRNA on the bone tissue.
