摘 要:目的探究硫氧还蛋白9(TXNDC9)在肝细胞肝癌(HCC)组织中的表达及其与TACE治疗疗效的关系。方法选择2013年1月至2016年1月诊断为HCC的97例患者作为研究对象。用改良实体瘤疗效评价标准(mRECIST)评价TACE疗效。用免疫印迹法检测HCC组织中TXNDC9水平,并分析其与HCC患者TACE治疗疗效的关系。结果完全缓解(CR)组(17例)、部分缓解(PR)组(30例)、疾病稳定(SD)组(21例)和疾病进展(PD)组(29例)HCC组织中TXNDC9水平分别为(0.66±0.09)、(0.75±0.09)、(0.94±0.12)和(1.08±0.11),差异有统计学意义(P<0.05)。HCC组织中TXNDC9水平与TACE治疗疗效呈负相关(P<0.05)。TXNDC9诊断HCC预后的受试者工作特征(ROC)曲线下面积(AUC)、灵敏度和特异度分别为0.930、73.91%和96.08%。多因素Cox回归分析结果显示肿瘤直径、Child-Pugh分级和TXNDC9是HCC患者预后的独立影像因素(P<0.05)。结论HCC组织中TXNDC9水平高提示TACE治疗疗效较差,预后不良风险较高。Objective To investigate the correlation between the expression level of thioredoxin domain containing protein 9(TXNDC9)in hepatocellular carcinoma(HCC)tissues and the curative efficacy of transcatheter arterial chemoembolization(TACE).Methods A total of 97 patients diagnosed as HCC from January 2013 to January 2016 were selected as the subjects of this study.Modified response evaluation criteria in solid tumor(m RECIST)was used to evaluate the efficacy of TACE treatment.The level of TXNDC9 in HCC tissues was detected by Western blotting,and its relationship to the curative efficacy of TACE treatment in HCC patients was analyzed.Results The levels of TXNDC9 in HCC tissues of complete remission(CR)group(n=17),partial remission(PR)group(n=30),stable disease(SD)group(n=21)and progression disease(PD)group(n=29)were(0.66±0.09),(0.75±0.09),(0.94±0.12)and(1.08±0.11),respectively,and the differences were statistically significant(P<0.05).A negative correlation existed between the TXNDC9 level in HCC tissues and the curative effect of TACE treatment(P<0.05).The area under the receiver operating characteristic(ROC)curve,the sensitivity and the specificity of TXNDC9 in predicting HCC prognosis were 0.930,73.91%and 96.08%,respectively.Multivariate Cox regression analysis showed that tumor diameter,Child-Pugh classification and TXNDC9 level were the independent factors affecting prognosis in HCC patients(P<0.05).Conclusion A high level of TXNDC9 in HCC tissues indicates a poor curative effect of TACE and a high risk of poor prognosis.
