摘 要:目的探讨脑源性神经营养因子(BDNF)与中枢性肥胖抵抗的相关性。方法将新生健康小鼠随机分为对照组、实验组和模型组。实验组和模型组自小鼠出生当天起,按3 mg·g^(-1)·d^(-1)的剂量颈部皮下注射10%谷氨酸钠(MSG),连续5 d;对照组同法注射等剂量0.9%NaCl。根据实验要求对小鼠进行剔除,最终对照组、实验组和模型组各8只小鼠。比较各组小鼠体重、体长、Lee’s指数、血清中血脂含量,棕色脂肪(BAT)和肾周白色脂肪(WAT)重量变化;用蛋白质印迹(Western blotting)法检测脂肪组织中BDNF的表达水平。结果对照组、模型组和实验组的体重分别为(40.62±3.94),(28.54±4.77),(48.89±3.15)g,三酰甘油(TG)分别为(0.73±0.35),(0.97±0.35),(1.40±0.52)mmol·L^(-1),总胆固醇(CHOL)分别为(1.97±0.45),(1.49±0.45),(2.94±0.18)mmol·L^(-1),高密度脂蛋白(HDL)分别为(1.97±0.41),(1.60±0.51),(2.76±0.17)mmol·L^(-1),低密度脂蛋白(LDL)分别为(0.24±0.02),(0.24±0.10),(0.32±0.04)mmol·L^(-1),白色脂肪重量分别为(0.20±0.06),(0.28±0.14),(0.80±0.24)g,棕色脂肪重量分别为(0.12±0.02),(0.20±0.10),(0.36±0.09)g,实验组与对照组和模型组比较,以上指标差异均有统计学意义(均P<0.05)。结论脂肪组织中BDNF表达上调与中枢性肥胖抵抗存在密切相关性。Objective To investigate the relationship between brain-derived neurotrophic factor(BDNF) and entral obesity-resistant. Methods Healthy kunming mice at the neonatal stage were randomly divided into control group, experimental group and model group. To obtain the obese models, experimental group and model group mice were injected at cervical subcutaneous with 10% monosodium glutamate(MSG)3 mg·g^(-1)·d^(-1) for 5 d. Control group was injected with the same dose of 0.9% NaCl. In addition, mice were removed according to the requirements. Finally, 8 mice in each group. The body weight, body length, Lee’s index, brown fat(BAT), perirenal white fat(WAT) weight, and levels of serum lipid were compared in each group, the BDNF expression levels in adipose tissue were measured by Western blotting. Compared with control group and model group,the above indexes in experimental group were all with significant difference( all P < 0. 05). Conclusion Up-regulation of BDNF in adipose tissue may be involved with central obesity-resistant of mice.
