摘 要:目的观察双夏汤对肝郁型失眠大鼠行为学及脑内单胺类神经递质的作用。方法将70只雄性Wistar大鼠随机分为正常组、模型组、安定组、泻肝安神丸组及双夏汤高、中、低剂量组,每组10只。除正常组外,其余组均采用束缚和夹尾激怒轮流刺激叠加对氯苯丙氨酸(PCPA)致肝郁型失眠模型。造模第8天,安定组给予4.2 mg/kg安定灌胃,泻肝安神丸组给予1.4 g/kg泻肝安神丸灌胃,双夏汤高、中、低剂量组分别给予7 g生药/kg、3.5 g生药/kg、1.7 g生药/kg双夏汤灌胃,正常组和模型组给予等量蒸馏水灌胃,均每天1次,连续2周。观察各组大鼠的体征、体重及进食量,大鼠在敞箱中自主活动情况,戊巴比妥钠诱导睡眠的入睡潜伏期和睡眠时间,并用高效液相色谱-电化学检测法测定各组大鼠脑组织中去甲肾上腺素(NE)、多巴胺(DA)、5-羟吲哚乙酸(5-HIAA)、5-羟色胺(5-HT)含量。结果模型组大鼠造模第7,14,21天的体重、进食量、水平运动和垂直运动得分均明显低于正常组(P均<0.05);造模第21天,双夏汤高剂量组大鼠的体重、进食量和双夏汤中、低剂量组大鼠的水平运动和垂直运动得分均明显高于同期模型组(P均<0.05),其余组各项指标与同期模型组比较差异均无统计学意义(P均>0.05)。模型组大鼠入睡潜伏期明显长于正常组(P<0.05),各药物组大鼠的入睡潜伏期均明显短于模型组(P均<0.05);安定组和双夏汤高剂量组大鼠睡眠时间均明显长于模型组(P均<0.05),其余组大鼠睡眠时间与模型组比较差异均无统计学意义(P均>0.05)。大脑皮质中,模型组大鼠NE、DA、5-HIA、5-HT含量及5-HT/NE、5-HT/DA均明显低于正常组(P均<0.05),各给药组大鼠各指标与模型组比较差异均无统计学意义(P均>0.05);下丘脑中,模型组大鼠DA、5-HIAA、5-HT含量及5-HT/NE、5-HT/DA均明显低于正常组(P均<0.05);双夏汤高剂量组和双夏汤低剂量组NE、5-HT含量及5-HT/Objective It is to observe the effects of Shuangxia Decoction on the behavior and brain monoamine neurotransmitters in rats with insomnia of type of liver depression.Methods Seventy male Wistar rats were randomly divided into normal group,model group,diazepam group,Xiegan Anshen pill group and high,medium and low dose groups of Shuangxia Decoction,each group had 10 rats.Except for the normal group,the rest groups were alternately stimulated with blinding and tail-clamping irritation plus parachlorophenylalanine(PCPA)to induce the models of insomnia with liver depression.On the 8th day of model building,the diazepam group was given 4.2 mg/kg diazepam by gavage,the Xiegan Anshen pill group was given 1.4 g/kg Xiegan Anshen pill by gavage,and the high,medium and low dose groups of Shuangxia Decoction were given 7 g crude drug/kg,3.5 g crude drug/kg,1.7 g crude drug/kg Shuangxia Decoction by gavage,the normal group and the model group were given the same amount of distilled water by gavage,all once a day and treated for 2 weeks.The physical signs,body weight and food intake,the autonomous activity in the open box,the latency and sleep time of pentobarbital sodium-induced sleep of the rats in each group were observed,and the content of norepinephrine(NE),dopamine(DA),5-hydroxyindole acetic acid(5-HIAA)and serotonin(5-HT)in rat brain tissue were determined by high performance liquid chromatography-electrochemical detection method.Results The weight,food intake,horizontal movement and vertical movement scores of the rats in the model group were significantly lower than those of the normal group on the 7,14,and 21 days(all P<0.05);on the 21st day,the body weight,food intake of rats in the high-dose Shuangxia Decoction group and the horizontal and vertical movement scores of rats in the medium and low dose groups of Shuangxia Decoction were significantly higher than those of the model group in the same period(all P<0.05),there was no statistically significant difference in the indicators of the other groups compared with t
