摘 要:目的探讨丹参对脑缺血再灌注损伤脑组织中磷酸化AKT蛋白(p-AKT)、半胱氨酸蛋白酶-3(Caspase-3)和缺氧诱导因子-1α(HIF-1α)蛋白的影响。方法2020年3—5月,48只健康SD大鼠随机分为假手术组、模型组、丹参组和LY294002组,每组12只。丹参组灌胃给予丹参水提物灌胃,LY294002组肌内注射PI3K/AKT抑制剂LY294002,假手术组和模型组给予同体积生理盐水灌胃。采用线栓法建立局灶性脑缺血再灌注损伤模型。观察脑缺血再灌注后大鼠神经功能缺损情况及脑梗死体积,检测缺血脑组织中p-Akt、HIF-1α和Caspase-3蛋白水平的表达。结果造模后,假手术组神经功能评分、脑梗死体积均为0;LY294002组神经功能评分最高,脑梗死体积最大,其次为模型组,丹参组最低,组间比较差异有统计学意义(P<0.05)。造模后,丹参组HIF-1α水平最高,其次为模型组与LY294002组,均显著高于假手术组,差异有统计学意义(P<0.05);造模后,丹参组p-Akt水平最高,其次为模型组,LY294002组次之,均显著高于假手术组,差异有统计学意义(P<0.05);造模后,LY294002组Caspase3水平最高,其次为模型组,丹参组次之,均显著高于假手术组,差异有统计学意义(P<0.05)。结论丹参对脑缺血再灌注损伤具有保护作用,其机制可能与激活PI3K/Akt信号通路,上调p-Akt和HIF-1α蛋白表达,降低Caspase-3表达有关。Objective To investigate the impacts of salvia miltiorrhiza on phosphorylated AKT protein(p-AKT),cysteine proteinase-3(Caspase-3)and Hypoxia inducible factor-1α(HIF-1α)proteins in brain tissue of cerebral ischemia-reperfusion injury.Methods A total of 48 healthy SD rats from March to May 2020 were randomly divided into the sham operation group(n=12),the model group(n=12),the salvia miltiorrhiza group(n=12)and the LY294002 group(n=12).Rats in the salvia miltiorrhiza group were treated with water extract of salvia miltiorrhiza by gavage,rats in the LY294002 group were treated with PI3K/AKT inhibitor LY294002 intramuscularly,and rats in the sham operation group and the model group were treated with the same volume of normal saline by gavage.The model of focal cerebral ischemia-reperfusion injury was established by suture method.The neurological deficit and cerebral infarction volume after cerebral ischemia-reperfusion in rats were observed,and the levels of p-AKT,HIF-1αand Caspase-3 protein expression in ischemic brain tissue were detected.Results After modeling,the neurological function score and cerebral infarction volume of the sham operation group were all 0.There was the highest neurological function score and the largest cerebral infarction volume in the LY294002 group.The model group was the second,and the salvia miltiorrhiza group was the lowest,with statistically significant difference among the groups(P<0.05).After modeling,the level of HIF-1αin the salvia miltiorrhiza group was the highest,and the model group and the LY294002 group were the second.All of them were significantly higher than that in the sham operation group,with statistically significant difference(P<0.05).After modeling,the level of p-AKT in the salvia miltiorrhiza group was the highest,the model group was the second,and the LY294002 group was the third,which were all significantly higher than that in the sham operation group,with statistically significant difference(P<0.05).After modeling,the Caspase-3 level in the LY294002 group was
