补阳还五汤对脑出血大鼠脑组织Ang-1和Tie-2表达的影响

摘　　要：目的探讨补阳还五汤对脑出血大鼠脑组织中促血管生成素1(Ang-1)和酪氨酸激酶受体2(Tie-2)表达的影响。方法96只SD大鼠随机分为三组,每组32只。A组和M组注射胶原蛋白酶Ⅶ构建大鼠脑出血模型,S组为假手术组。A组每日采用补阳还五汤2 mL灌胃,每天2次,连续治疗14 d。A组和M组在脑出血建模后第1、3、7、14天获取大鼠出血部位脑组织,实时荧光定量PCR检测脑组织中Ang-1和Tie-2 mRNA表达,分析其相关性。结果M组Ang-1和Tie-2 mRNA表达水平在第1天低于S组(P<0.01),但第7、14天高于S组(P<0.01)。第3、7、14天,A组Ang-1和Tie-2 mRNA表达水平均高于M组(P<0.05或P<0.01)。Ang-1与Tie-2 mRNA表达水平呈正相关(r=0.89,P<0.01)。结论脑出血大鼠脑组织中Ang-1和Tie-2 mRNA表达水平先下降后升高,补阳还五汤可能通过上调Ang-1和Tie-2 mRNA表达发挥治疗脑出血的作用。Objective To investigate the effect of Buyang Huanwu decoction on the expressions of angiopoietin 1(Ang-1)and tyrosine kinase receptors 2(Tie-2)in brain tissues of the rats with cerebral hemorrhage.Methods Ninety-six SD rats were randomly divided into three groups with 32 rats each.The rat models with cerebral hemorrhage were established by collagen enzymeⅦinjection in groups of A and M.Group S was taken as the sham operation control.The rats in group A were treated with Buyang Huanwu decoction 2 mL gastric gavage twice per day for 14 days after establishment of cerebral hemorrhage.Brain tissues from the site of cerebral hemorrhage of group A and group M were obtained on the 1st,3rd,7th and 14th day for the detection of Ang-1 and Tie-2 mRNA expressions by real-time fluorescence quantitative PCR.The correlation between Ang-1 and Tie-2 mRNA expressions was analyzed.Results The mRNA expression levels of Ang-1 and Tie-2 on the 1st day in group M were lower than those in group S(P<0.01),which were higher than those in group S on the 7 th day and 14th day(P<0.01).Ang-1 expression was positively correlated with Tie-2 mRNA expression(r=0.89,P<0.01).Conclusion The mRNA expression levels of Ang-1 and Tie2 in brain tissues of the rats with cerebral hemorrhage are decreased firstly and then increased.Buyang Huanwu decoction may play a role in the treatment of cerebral hemorrhage by up-regulating the mRNA expression levels of Ang-1 and Tie-2.
