摘 要:目的探讨宫颈癌患者手术前后血清前梯度蛋白2 (AGR2)、结缔组织生长因子(CTGF)水平变化,分析二者与临床病理参数的关系,为宫颈癌辅助诊断和治疗提供参考依据。方法选取2014年3月-2018年4月成都市第二人民医院收治的65例宫颈癌患者为宫颈癌组,同时选取65例来该院体检的健康志愿者为正常对照组。采用ELISA检测两组血清AGR2和CTGF水平,分析二者和宫颈癌患者临床病理特征的关系,采用Cox逐步回归分析宫颈癌的影响因素。结果宫颈癌组患者术前术后血清中AGR2表达水平高于正常对照组,术后血清AGR2表达水平较术前下降(均P<0.05);宫颈癌组患者术前术后血清CTGF表达水平低于正常对照组,术后CTGF水平较术前升高(均P<0.05)。宫颈癌组患者术前血清AGR2、CTGF水平与肿瘤大小、FIGO分期、淋巴结转移有关。Cox逐步回归分析显示,FIGO分期II期、伴有淋巴结转移、AGR2高表达、CTGF低表达为宫颈癌的独立危险因素(P<0.05)。结论宫颈癌患者血清AGR2、CTGF存在异常表达,二者表达水平均与宫颈癌患者的肿瘤大小、FIGO分期、淋巴结转移有关,AGR2高表达、CTGF低表达为宫颈癌的独立危险因素,可能为宫颈癌的治疗提供新靶点。Objective To explore the changes of expression levels of serum anterior gradient protein 2( AGR2) and connective tissue growth factor( CTGF) in patients with cervical cancer before and after operation,analyze the correlations with clinicopathological parameters,and provide a certain reference basis for auxiliary diagnosis and treatment of patients with cervical cancer. Methods From March 2014 to April2018,65 patients with cervical cancer treated in Chengdu Second People’s Hospital were selected as cervical cancer group,65 healthy volunteers were selected as normal control group. ELISA was used to detect the expression levels of serum AGR2 and CTGF in the two groups. The correlations with clinicopathological parameters were analyzed. Cox stepwise regression analysis was used to analyze the influencing factors of cervical cancer. Results The expression levels of serum AGR2 before and after operation in cervical cancer group were higher than those in normal control group,the expression level of serum AGR2 after operation in cervical cancer group was lower than that before operation( P<0. 05). The expression levels of serum CTGF before and after operation in cervical cancer group were lower than those in normal control group,the expression level of serum CTGF after operation in cervical cancer group was higher than that before operation( P<0. 05). The levels of serum AGR2 and CTGF before operation in cervical cancer group were correlated with tumor size,FIGO stage,and lymph node metastasis. Cox stepwise regression analysis showed that FIGO stageⅡ,lymph node metastasis,high expression of AGR2,and low expression of CTGF were independent risk factors of cervical cancer( P<0. 05). Conclusion Abnormal expression levels of serum AGR2 and CTGF in patients with cervical cancer are related to tumor size,FIGO stage,and lymph node metastasis. High expression of AGR2 and low expression of CTGF are independent risk factors of cervical cancer,which may provide a new target for treatment of cervical cancer.
