摘　　要：目的:探讨FAP-1在胶质母细胞瘤(glioblastoma,GBM)组织中的表达及其对胶质瘤细胞增殖、迁移作用的影响。方法:通过Oncomine数据库探讨FAP-1在胶质瘤中的表达,并通过PCR测定胶质母细胞瘤组织及非胶质母细胞瘤组织中FAP-1的表达水平;通过不同浓度FAP-1 siRNA转染胶质瘤细胞系LN18、U87,降低FAP-1表达,通过吸光度实验观察FAP-1表达对胶质母细胞瘤细胞系增殖能力影响,划痕实验评估FAP-1对胶质瘤细胞系迁移能力影响。结果:MTT实验提示沉默FAP-1表达促进胶质母细胞瘤细胞增殖能力(P<0.05)。划痕实验提示沉默FAP-1表达则促进胶质母细胞瘤细胞迁移能力。结论:FAP-1在胶质母细胞瘤中可能发挥着抑癌基因的功能。Objective:To investigate the expression and function of FAP-1 in glioblastoma and its effect on proliferation and metastasis of glioma cells.Methods:Oncomine database was used to examine the expression of FAP-1 in GBM and then PCR was performed to test the results,different concentration of FAP-1 siRNA targeted FAP-1 were applied to adjust the expression levels of FAP-1,scratch test and absorbance test demonstrate were performed to test the effect of FAP-1 on cell proliferation and migration.Results:MTT and scratch test suggested that knocked down of FAP-1 resulted in enhancement of cell proliferation and migration.Conclusion:FAP-1 may play a role of cancer suppressor gene in GBM.
