摘  要:肌内脂肪(Intramuscular fat,IMF)是重要的风味物质,是影响鸡肉品质的重要因素之一。IMF可以切断肌纤维束间的交联结构,同时对咀嚼过程中的肌纤维断裂有利,从而提高肉的嫩度。为探究IMF含量与过氧化物酶体增殖物激活受体γ(Peroxisome proliferator activatedreceptorγ,PPARγ)基因及肝脏脂肪酸结合蛋白(Fatty acid-binding protein,L-FABP)基因的表达量关系,试验以镇沅瓢鸡和盐津乌骨鸡为研究对象,在相同饲养条件下于300日龄进行屠宰(公母各半),分别测定肌肉组织中IMF含量以及PPARγmRNA和L-FABP mRNA表达量,并进行相关性分析。结果显示:①镇沅瓢鸡胸肌和腿肌IMF含量显著高于盐津乌骨鸡胸肌和腿肌;两种鸡腿部肌肉IMF含量均显著高于胸部肌肉。②不同鸡种间显示,盐津乌骨鸡胸肌中PPARγmRNA及L-FABP mRNA表达水平高于镇沅瓢鸡,差异极显著(P<0.01);镇沅瓢鸡腿肌中PPARγmRNA及表达水平低于盐津乌骨鸡,差异显著(P<0.05),而L-FABP mRNA表达水平极显著高于盐津乌骨鸡(P<0.01)。研究结果表明:相同饲养条件下的镇沅瓢鸡肉品质优于盐津乌骨鸡,且PPARγ基因及L-FABP基因对肌内脂肪的合成起关键作用。Intramuscular fat(IMF) is an important flavour substance and one of the important factors affecting meat quality. IMF can cut the cross-linked structure between muscle fibre bundles, and beneficial to muscle fibre breakage during chewing, thereby improving meat tenderness. To explore the relationship between the content of IMF and the expression of peroxisome proliferator-activated receptor γ(PPARγ) gene and fatty acid-binding protein(L-FABP) gene,Piao chicken and Yanjin silky chicken were slaughtered at 300 days of age under the same feeding conditions(male and female half each), and the content of IMF in muscle tissue and the expression of PPARγ mRNA and L-FABP mRNA were measured. The results showed:(1) The IMF content of Piao chicken breast and leg muscles was significantly higher than that of Yanjin silky chicken;the IMF content of the leg muscles of the two chickens were significantly higher than that of the breast muscles, and the female IMF content was both in the breast muscles and the legs.(2) Different chicken breeds showed that the expression levels of PPARγ mRNA and L-FABP mRNA in the breast muscles of Yanjin silky chickens were higher than those of Piao chickens, and the difference was extremely significant(P<0.01);PPARγ mRNA and expression levels in the leg muscles of Piao chickens was higher than that of Yanjin silky chickens, the difference was significant(P<0.05), and the expression level of L-FABP mRNA was extremely significantly higher than that of Yanjin silky chickens(P<0.01). The results indicated that the meat quality of Piao chicken under the same feeding conditions was better than that of Yanjin Silky chicken, and the PPARγ and L-FABP genes played a role in the synthesis of IMF.
