摘 要:采用乙醚连续渗漉提取白秃马勃粗脂肪,利用硅胶柱色谱对白秃马勃粗脂肪化学成分进行分离纯化,通过气相色谱-质谱联用技术(GC-MS)分析菌油脂肪酸成分,运用电喷雾电离质谱(ESI-MS)、一维核磁共振谱(1D NMR)和二维核磁共振谱(2D NMR)解析化合物结构,通过体外实验测试菌油对1,1-二苯基-2-苦基肼基(DPPH)的清除能力。结果表明,白秃马勃菌油主要脂肪酸为亚油酸(44.11%)、油酸(27.49%)和棕榈酸(17.60%)。2个单体化合物被解析为麦角甾-5,8-表二氧-6,22-二烯-3-乙酸酯(MB1)和麦角甾-5,8-表二氧-6,22-二烯-3-醇(MB2)。白秃马勃菌油在试验浓度范围内具有一定的DPPH自由基清除能力,半数清除浓度(IC50)为26.61 mg/mL,并且呈浓度依赖关系。化合物MB1和MB2均为首次从白秃马勃菌分离得到。The crude fat of the puffball Calvatia candida(Rostk.)Hollós was extracted by ether and purified by silica column chromatography.The fatty acid composition of the fungal oil was analyzed by gas chromatography-mass spectrometry(GC-MS),and the compound’s structure was elucidated by electrospray ionization mass spectrometry(ESI-MS),one-dimensional nuclear magnetic resonance spectroscopy(1 D NMR)and two-dimensional nuclear magnetic resonance spectroscopy(2 D NMR),and the scavenging activity of the fungal oil against 1,1-diphenyl-2-bitter hydrazine(DPPH)was tested in vitro.The results show that the main fatty acids in the fungal oil were linoleic acid(44.11%),oleic acid(27.49%)and palmitic acid(17.60%),respectively.The two monomer compounds were elucidated as ergosta-5,8-phenylenedio-6,22-diene-3-yl acetate(MB1)and ergosta-5,8-phenylenedio-6,22-diene-3-ol(MB2),respectively.The fungal oil had DPPH scavenging activity in the range of experimental concentration with the half scavenging concentration(IC50)of 26.61 mg/mL,and showed concentration dependence.Both the compounds MB1 and MB2 were isolated from C.candida for the first time.
