摘  要:为阐明梨网蝽Stephanitis nashi对其寄主植物的化学感受机制,本研究采用高通量测序平台对梨网蝽成虫触角进行转录组测序,基于7大数据库对转录组数据进行基因功能注释,筛选获得梨网蝽的主要嗅觉相关基因并进行生物信息学和系统进化树分析。结果表明:共获得28017条unigenes,长度在1000 bp以上的unigenes有9921条;通过与7大数据库进行BLAST比对,梨网蝽触角转录组数据中的17891条unigenes被成功注释;在已成功注释的unigenes中,鉴定得到12个OBP基因和17个CSP基因,其中OBP基因所编码的蛋白有6个属于Classic家族;而CSP基因所编码的蛋白全部具有4个保守的半胱氨酸位点,符合昆虫CSP基因的结构特征。进化分析结果显示,梨网蝽6个OBP基因的氨基酸序列与其他昆虫OBP基因的氨基酸序列聚为3个分支;梨网蝽16个CSP基因与其他昆虫CSP基因的进化树也聚为3个分支。In order to clarify the chemosensory mechanism of pear lace bug Stephanitis nashi to orientate its host plant,the antennal transcriptome of adults S.nashi was sequenced using the Illumina Hiseq 4000 platform,annotated based on the seven major data bases,and the main olfaction-related genes of S.nashi were screened out and further analyzed.The results of bioinformatic analysis showed that there were 28017 unigenes,among which the nucleotide sequence lengths of 9921 unigenes were over one kb in size.A total of 17891 unigenes in the antennal transcriptome of S.nashi adults were successfully annotated using BLAST search.Through further transcriptome data analysis,12 OBP genes and 17 CSP genes were identified.Among them,six proteins encoded by OBP genes belonged to the classic family,and all of the proteins encoded by CSP genes had four conserved cysteine sites,consistent with the structural characteristics of insect CSP genes.The results of phylogenetic analysis showed that the amino acid sequences of six OBP genes of Stephanitis and other insect OBP genes were clustered into three branches,and the evolutionary trees of 16 CSP genes and other insect CSP genes were also divided into three branches.
