摘  要:脑胶质瘤是中枢神经系统最为常见的原发性肿瘤,其中胶质母细胞瘤(glioblastoma multiforme,GBM)发病率较高,其治疗效果主要依赖于手术切除程度及术后化疗效果。由于颅内血脑屏障的作用和耐药基因O~6-甲基鸟嘌呤-DNA甲基转移酶的产生,使得化疗药物在脑胶质瘤患者中的作用大大受限,难以达到预期的临床效果。因此,对脑胶质瘤耐药机制的深入研究意义重大。随着近年来对核转录因子κB(nuclear factor kappa B,NF-κB)通路的深入研究,证明该通路的激活可促进肿瘤细胞扩散和诱导耐药,若拮抗NF-κB通路上下游信号分子可显著改善耐药现象。本文就近年来NF-κB的研究进展以及NF-κB如何参与胶质瘤耐药作一综述。Glioma is the most common primary tumor of thecentral nervous system,and the incidence of glioblastoma multiforme(GBM)is high.The treatment effects mainly depend on the degree of surgical resection and postoperative chemotherapy.Due to effects of intracranial blood-brain barrier and the generation of drug-resistant gene O~6-methylguanine-DNA methyltransferase(MGMT),the effectiveness of chemotherapeutics for glioma patients is greatly limited,and it is difficult to achieve expected clinical results.Therefore,it is important to study the mechanism of drug resistance of glioma.In recent years,deep researches on nuclear factor kappa B(NF-κB)pathway have proved that activation of this pathway can promote the proliferation of tumor cells and induce drug resistance,and antagonizing the upstream and downstream signaling molecules of NF-κB pathway can significantly improve drug resistance.In this review,the research progress of NF-κB in recent years and the role of NF-κB in glioma drug resistance will be summarized.
