前列腺癌中CD147、MMP-2 mRNA表达与临床分期、病理学分级的关系

摘　　要：目的分析前列腺癌病灶组织中细胞外基质金属蛋白酶诱导因子CD147、基质金属蛋白酶(MMP)-2(MMP-2)mRNA表达与临床分期、病理学分级的关系。方法选取2015年4月至2018年12月本院收治的前列腺癌患者58例及同期行手术治疗的前列腺增生患者50例为研究对象,取前列腺癌患者病灶组织、癌旁组织及前列腺增生患者的组织标本,以原位杂交法测定各组织中CD147 mRNA表达水平,采用逆转录-聚合酶链反应(RT-PCR)测定MMP-2 mRNA,并评估CD147 mRNA表达与临床分期、病理学分级等的关系。结果前列腺癌病灶组织中CD147 mRNA阳性率及MMP-2表达水平高于前列腺癌癌旁组织、前列腺增生组织(P<0.05),前列腺癌癌旁组织CD147 mRNA阳性率及MMP-2表达水平也高于前列腺增生组织(P<0.05);前列腺癌患者中,临床分期T3~T4期、病理学分级≥5级、淋巴结转移N1期、远处转移M1期者的CD147 mRNA阳性率、MMP-2 mRNA表达水平高于临床分期T1~T2期、病理学分级2~4级、淋巴结转移N0期、远处转移M0期者,差异有统计学意义(P<0.05)。结论检测CD147、MMP-2 mRNA的表达对评估肿瘤恶性程度与生物学行为、预后有积极意义。Objective To analyze the relationship between expression of(Extracellular Matrix Metalloproteinase Induce)CD147,matrix metalloproteinase-2(MMP-2)mRNA and clinical staging,pathological grading in tissues from prostate cancer(PCA).Methods A total of 58 PCA patients who were admitted to the First Clinical Medical College of Guangzhou University of Traditional Chinese Medicine from April2015 to December 2018 and 50 patients with benign prostatic hyperplasia(BPH)who underwent surgery in the same period were enrolled in the study.The tissues and adjacent tissues of PCA patients,and tissue specimens of BPH patients were collected.The expression levels of CD147 mRNA in all tissues were measured by in situ hybridization.The reverse transcription-polymerase chain reaction(RT-PCR)was performed to measure MMP-2 mRNA.The relationship between expression of CD147 mRNA and clinical staging,pathological grading was evaluated.Results The positive rate of CD147 mRNA and MMP-2 expression level in PCA tissues were higher than those in PCA adjacent tissues and BPH tissues(P<0.05).The positive rate of CD147 mRNA and MMP-2 expression level in PCA adjacent tissues were higher than those in BPH tissues(P<0.05).Among PCA patients,the positive rates of CD147 m RNA and MMP-2 expression level in patients with clinical stage at T3 to T4 stage,pathological grade of greater than 5 level,lymph node metastasis at N1 stage and distant metastasis at M1 stage were higher than those with clinical stage at T1 to T2 stage,pathological grade within 2 to 4 level,lymph node metastasis at N0 stage and distant metastasis at M0 stage(P<0.05).Conclusion The expression of CD147 and MMP-2 mRNA in PCA tissues is closely related to the clinical staging and pathological grading.The detection of both CD147 and MMP-2 mRNA is of positive significance for evaluating the tumor malignancy degree,biological behavior,and prognosis.
