摘  要:目的观察丁苯酞给药时间差异对SD大鼠大脑中动脉脑梗死模型神经轴突生长抑制剂A(Nogo-A)、血管内皮生长因子(VEGF)、生长相关蛋白43(GAP43)表达及神经功能恢复的影响。方法选择SPF级雄性SD大鼠18只,建立大脑中动脉脑梗死模型,随机分为对照组、即刻给药组、延迟给药组,每组6只。检测大鼠术前,术后3 h、48 h、7 d、14 d血清Nogo-A和14 d脑组织Nogo-A水平,监测术前和术后3 h、48 h、7 d、14 d Ludmila Belayev 12评分,14 d后处死大鼠,比较各组脑梗死体积,镜检脑梗死病灶和对侧正常脑组织苏木精-伊红染色和VEGF、GAP43表达情况。结果即刻给药组和延迟给药组脑梗死体积比明显低于对照组(0.117±0.011、0.185±0.010 vs 0.356±0.140,P<0.01),且即刻给药组脑梗死体积比明显低于延迟给药组(P<0.01)。即刻给药组48 h、7 d和14 d血清Nogo-A水平明显低于对照组,延迟给药组7 d和14 d血清Nogo-A水平明显低于对照组,延迟给药组48 h血清Nogo-A水平明显高于即刻给药组(P<0.01)。即刻给药组术后7 d和14 d Ludmila Belayev 12评分明显低于对照组,延迟给药组术后7 d和14 d Ludmila Belayev 12评分明显高于即刻给药组(P<0.05)。Pearson相关性分析显示,术后48 h、7 d和14 d血清Nogo-A水平与Ludmila Belayev 12评分相关(P<0.05)。结论丁苯酞即刻给药和延迟给药均可改善脑卒中大鼠神经功能预后,并对Nogo-A、VEGF和GAP43表达有显著影响。Objective To study the effect of butylphthalide administration at differen time points on expressions of Nogo-A,VEGF,GAP43 and neurological function recovery in middle cerebral artery cerebral infarction model of SD rats.Methods Eighteen SD rats were randomly divided into control group(n=6),immediate butylphthalide administration group(n=6)and delayed butylphthalide administration group(n=6).A middle cerebral artery infarction model of rats was established.The serum Nogo-A level in different groups was measured before operation,at 3 and 48 h and on days 7 and 14 respectively after operation.The Ludmila Belayev score of different groups was recorded before operation,at 3 and 48 h and on days 7 and 14 respectively after operation.The animals were sacrificed.The volume of cerebral infarction in different groups was measured.The infarction lesion and contralateral normal brain tissue were observed under microscope with HE staining.The expressions of VEGF and GAP43 in different groups were detected.ResultsThe cerebral infarction volume was significantly smaller in immediate butylphthalide administration group and delayed butylphthalide administration group than in control group(0.117±0.011,0.185±0.010 vs 0.356±0.140,P<0.01)and in immediate butylphthalide administration group than in delayed butylphthalide administration group(P<0.01).The serum Nogo-A level was significantly lower in immediate butylphthalide administration group than in control group at 48 h and on days 7 and 14 after operation and in delayed butylphthalide administration group than in control group on days 7 and 14 after operation(P<0.01)and was significantly higher in delayed butylphthalide administration group than in immediate butylphthalide administration group at 48 h after operation(P<0.01).The Ludmila Belayev 12 score was significantly lower in immediate butylphthalide administration than in control group on days 7 and 14 after operation and was significantly higher in delayed butylphthalide administration group than in immediate butylphthal
