摘 要:目的:研究口腔白斑发生发展中的血管生成基因表达谱,并验证关键基因表达。方法:对正常黏膜(n=3),低危白斑(n=4),高危白斑(n=4),早期鳞癌(n=6)进行转录组学芯片检测生物学作用为血管生成的相关基因,对4个关键基因表达进行了验证。结果:从高危白斑发展到鳞癌有151个基因,为全部异常表达基因占比18.9%。RT-qPCR验证结果显示VEGFA、PDGFRB、MMP1和MMP3在正常黏膜、口腔白斑和鳞癌中的表达量均呈阶梯式升高,与芯片结果基本一致。结论:血管生成基因的异常表达与白斑癌变关系密切。Objective:To study the expression profile of angiogenic genes in the development of oral leucoplakia(OLK)and to verify the expression of key genes.Methods:The angiogenic genes of OLK in normal mucosa(n=3),low risk OLK(n=4),high risk OLK(n=4)and early OSCC(n=6)samples were detected by transeriprome microarray and the expression of 4 key genes were verified by RT-qPCR.Results:There were 151 differentially expressed genes from high risk OLK to OSCC,accounted for 18.9%of all abnormal genes.qRT-PCR results showed that the expression levels of VEGFA,PDGFRB,MMP1 and MMP3 in normal mucosa,OLK and OSCC increased in turn and inkeep with the results of microarray.Conclusion:Abnormal expression of angiogenic genes is closely related to oral carcinogenesis.
