摘　　要：目的探讨长链非编码RNA LSAMP-AS1在人胃癌组织中的表达及临床意义。方法收集2016年1月至2018年6月手术治疗60例患者胃癌组织标本。采用实时荧光定量聚合酶链反应法(RT-qPCR)检测4株胃癌细胞系和1株正常胃黏膜细胞细胞系LSAMP-AS1的表达水平。结果LSAMP-AS1在胃癌组织中表达量明显低于癌旁组织。LSAMP-AS1表达量与淋巴结是否转移、TNM分期、BMI相关。多因素Logistic回归分析,胃癌转移与LSAMP-AS1表达水平相关。LSAMP-AS1组织表达水平对胃癌诊断的特异性为60.00%,敏感性为60.00%(AUC=0.607,P=0.043)。结论LSAMP-AS1可能作为抑癌基因参与胃癌的发生、发展,特别是参与胃癌的侵袭、转移过程,低表达LSAMP-AS1可作为不良预后指标,为胃癌诊断、预后判断提供重要参考依据。Objective To investigate the expression and clinical significance of LncRNA LSAMP-AS1 in human gastric cancer.Methods 60 samples were collected from patients with gastric cancer during January 2016 to June 2018 in the Affiliated Hospital of North Sichuan Medical College.Real-time quantitative polymerase chain reaction(RT-qPCR)was used to detect the expression levels of LSAMP-AS1 in 4 gastric cancer cell lines and 1 normal gastric mucosal cell line.Results The expression level of LSAMP-AS1 in gastric cancer tissues was lower than that in adjacent tissues,and was correlated with lymph node metastasis,TNM stage and BMI;Multiple Logistic regression analysis showed that metastasis of gastric cancer was correlated with lSAMP-AS1 expression level;The specificity and sensitivity of lSAMP-AS1 tissue expression level in the diagnosis of gastric cancer were 60.00%and 60.00%(AUC=0.607,P=0.043),separately.Conclusion LSAMP-AS1 may be involved in the occurrence and development of gastric cancer as a tumor suppressor gene,especially in the invasion and metastasis of gastric cancer.Low expression of LSAMP-AS1 can be used as a poor prognostic indicator,providing an important reference basis for the diagnosis and prognosis of gastric cancer.
