摘 要:目的探讨白藜芦醇抑制胃癌细胞增殖和迁移的机制。方法CCK-8实验检测胃癌SGC-7901细胞株的增殖活性,RT-qPCR检测miRNA-491在SGC-7901细胞内的表达水平,Transwell实验检测各组SGC-7901细胞的迁移能力。结果随白藜芦醇浓度增加,SGC-7901细胞的增殖活力受到相应抑制,且白藜芦醇浓度>50μmol/L时,SGC-7901细胞的增殖活力受到显著抑制(P<0.05)。SGC-7901细胞经过白藜芦醇(50μmol/L)处理后,其迁移能力受到显著抑制(P<0.001);SGC-7901细胞经过白藜芦醇(50μmol/L)处理后,细胞内miR-491的表达量显著上调(P<0.001);转染miR-491 mimics脂质体,显著抑制SGC-7901细胞的增殖活力(P<0.05)和迁移能力(P<0.001)。结论白藜芦醇有可能通过上调miR-491的表达来抑制胃癌SGC-7901细胞的增殖和迁移。Objective To investigate the effects and action mechanism of resveratrol on proliferation and migration gastric cancer cell.Methods The proliferation activity of gastric cancer-SGC-7901 cell line was detected by CCK-8 assay.And RT-qPCR was used to detect the expression levels of miRNA-491 in SGC-7901 cells.Moreover the migration ability of SGC-7901 cells was detected by Transwell assay.Results With the increase of resveratrol concentration,the proliferation activity of SGC-7901 cells was inhibited,when the concentration of resveratrol was greater than 50μmol/L,the proliferation activity of SGC-7901 cells was significantly inhibited(P<0.05).After SGC-7901 cells were treated by resveratrol(50μmol/L),their migration ability was significantly inhibited(P<0.001).After SGC-7901 cells were treated by resveratrol(50μmol/L),the expression levels of miR-491 in the cells were significantly up-regulated(P<0.01).Moreover the transfection of miR-491 mimics liposomes significantly inhibited the proliferation activity and migration ability of SGC-7901 cells(P<0.05).Conclusion Resveratrol may inhibit the proliferation and migration of gastric cancer SGC-7901 cells by up-regulating the expression levels of miR-491.
