摘 要:目的:探讨猴头健胃灵片对功能性消化不良大鼠胃敏感性及5-HT分泌的影响。方法:30只雄性SD大鼠随机分为空白组、模型组、猴头健胃灵组,每组10只。模型组和猴头健胃灵组采用夹尾刺激法制备功能性消化不良大鼠模型。猴头健胃灵组予猴头健胃灵片水溶液(0.24 g/kg)灌胃,空白组、模型组予等体积纯水灌胃,1次/d。治疗7 d后苏木精-伊红染色观察大鼠胃组织形态;电子恒压器检测大鼠胃敏感性;ELISA法检测大鼠血清及胃黏膜5-HT含量。结果:(1)大鼠胃组织形态:模型组与空白组、猴头健胃灵组相比差异无统计学意义;(2)胃敏感性变化:与空白组比较,模型组胃敏感性显著升高(P<0.05);与模型组比较,猴头健胃灵组胃敏感性降低(P<0.05);(3)血清和胃5-HT含量变化:模型组大鼠血清和胃5-HT含量较空白组显著升高(P<0.05);与模型组比较,猴头健胃灵组大鼠血清和胃5-HT含量明显下降(P<0.05)。结论:猴头健胃灵片通过下调功能性消化不良模型大鼠血清和胃5-HT的表达,从而缓解胃内高敏状态,可能是其治疗功能性消化不良的作用机制之一。Objective: Effect of Houtou Jianweiling Pian(HTJWL) on gastric sensitivity and expression of 5-HT in stomach of rats with functional dyspepsia(FD). Methods: A total of 30 male Sprague-Dawley(SD) rats were randomly divided into blank group, model group and HTJWL group, with 10 rats in each group. The model group and the HTJWL group were administrated with tail clamping to establish FD models. The HTJWL group was given(concentration of 0.24 g/kg) HTJWL by gavage, while the model group and the blank group were given equal volume of pure water by gavage, once a day. After 7 days’ treatment, histomorphology of stomach in all groups was observed by HE staining, as well as gastric sensitivity was detected though barostat. 5-HT levels in serum and gastric mucosa were detected by ELISA. Results:(1)There was no significant difference on the gastric histomorphology of the model group and the HTJWL group, compared with the blank group.(2)The gastric sensitivity of the model group increased significantly compared with the blank group(P<0.05), and the gastric sensitivity of HTJWL group decreased significantly compared with the model group(P<0.05).(3)The content of 5-HT in serum and stomach of the model group increased significantly compared to the blank group(P<0.05), and the 5-HT content of HTJWL group decreased significantly compared with the model group(P<0.05). Conclusion: HTJWL can reduce the expression level of 5-HT in rats with functional dyspepsia and alleviating the gastric hypersensitivity, which may be one of the mechanisms for treating functional dyspepsia.
