摘  要:目的研究βB2晶体蛋白与小鼠年龄相关性白内障的发生相关性,并探讨βB2晶体蛋白与小鼠晶状体功能的相关性。方法通过胚胎打靶技术建立βB2晶体基因敲除的小鼠(βB2 KO组,n=50),以野生型(WT组,n=50)小鼠作为对照。通过紫外线照射晶状体诱导小鼠发生白内障,分别比较诱导白内障后4、8、12、16、20个月小鼠体重、晶状体重量和直径、白内障的发生率以及晶状体谷胱甘肽(GSH)含量。结果2组小鼠体重、晶状体的重量和直径、以及白内障的发生率均随年龄增长而增加,而晶状体GSH水平随年龄增加而降低。与WT组小鼠相比,同龄βB2 KO组小鼠体重差异无统计学意义(P>0.05)。与WT组小鼠相比,同龄βB2 KO组小鼠晶状体的重量均显著降低,差异均有统计学意义(P<0.05)。4~12月龄时2组小鼠晶状体的直径比较,差异无统计学意义(P>0.05);与WT组小鼠相比,16、20月龄时βB2 KO组小鼠晶状体直径均降低,差异均有统计学意义(P<0.05)。4、8月龄时2组小鼠晶状体GSH水平比较,差异无统计学意义(P>0.05);与WT组小鼠相比,12~20月龄时βB2 KO组小鼠晶状体GSH水平显著降低,差异均有统计学意义(P<0.05)。与WT组小鼠相比,4~12月龄时βB2 KO组小鼠白内障发生率显著升高,差异均有统计学意义(P<0.05);16、20月龄时2组小鼠白内障发生率比较,差异无统计学意义(P>0.05)。结论βB2晶体蛋白与小鼠年龄相关的白内障的发生有关,βB2基因敲除增加小鼠年龄相关的白内障的发生率,其机制可能与βB2基因敲除抑制晶状体发育有关。Objective To study the correlation betweenβB2 crystallin and the occurrence of age-related cataracts in mice,and to explore the correlation betweenβB2 crystallin and mouse lens function.Methods TheβB2 crystal gene knockout mice(βB2 KO group,n=50)were established by embryo targeting technology,and wild-type(WT group,n=50)mice were used as controls.Ultraviolet rays were used to irradiate the lens to induce cataracts at the age of 4,8,12,16,20 months mice.The weight,lens quality and volume,incidence of cataracts,and lens glutathione(GSH)content of mice of different ages were compared.Results The body weight,lens quality and volume,and the incidence of cataracts in the two groups of mice increased with age,while the lens GSH level decreased with age.Compared with mice in the WT group,there was no statistically significant difference in body weight in theβB2 KO group of the same age(P>0.05).Compared with the WT group,the weight of the lens in the same ageβB2 KO group were significantly reduced,and the differences were statistically significant(P<0.05).There was no statistically significant difference in the lens diameter of the two groups of mice between 4 to 12 months(P>0.05);compared with the WT group,the lens diameter of theβB2 KO group at the age of 16 and 20 months were reduced,the differences were statistically significant(P<0.05).There was no statistically significant difference in lens GSH levels between the two groups at the age of 4 and 8 months(P>0.05);compared with the WT group,the levels of GSH in the lens of theβB2 KO group at the age of 12 to 20 months were significantly reduced,the differences were statistically significant(P<0.05).Compared with mice in the WT group,the incidence of cataracts in theβB2 KO group at the age of 4 to 12 months were significantly increased,and the differences were statistically significant(P<0.05).There was no statistically significant difference in the incidence of cataract between the two groups of mice at the age of 16,20 months(P>0.05).ConclusionβB2 crystallin
